Role of nuclear receptors in ABC transporters expression

Abstract:

Tato bakalářská práce se věnuje jaderným receptorům a jejich účinkům na průběh buněčného cyklu a expresi transportních proteinů z rodiny ABC transportérů, především těch asociovaných s multilékovou rezistencí. Jaderné receptory patří mezi nejhojnější transkripční regulátory u živočichů i rostlin. Ovlivňují i transkripci ABC transportérů, což jsou membránové pumpy, které se účastní především exportu mnoha molekul, které nejsou schopny projít buněčnými membránami difůzí. V této práci je podán přehled vlivu jaderných receptorů na expresi ABC transportérů a na průběh buněčného cyklu a vlivu ABC transportérů na průběh buněčného cyklu. Na základě literární rešerše bylo prokázáno, že zmíněné interakce mezi třemi danými skupinami molekul opravdu existují. Součástí práce bylo také zjistit, zda existuje přímé funkční spojení mezi jadernými receptory, ABC transportéry a buněčným cyklem. Nalezená fakta existenci tohoto spojení podporují, ačkoliv mechanizmy těchto interakcí nejsou dosud zcela objasněny.

Abstract:

This thesis studies nuclear receptors and their effects on cell cycle progression and expression of ABC transporters family proteins, especially those associated with multidrug resistance. Nuclear receptors belong among the most abundant transcriptional regulators in plants and animals. Nuclear receptors also affect transcription of ABC transporters. These transporters are membrane pumps, which especially participate in export of wide variety of molecules, which are not able to pass through biomembranes by diffusion. In this thesis is summarized the effect of nuclear receptors on cell cycle progression and on expression of ABC transporters, and the effect of ABC transporters on cell cycle progression. It has been established from literature, that there are interactions between these three above mentioned groups of molecules. One of the parts of this thesis is to reveal, whether there exists a functional interaction between nuclear receptors, ABC transporters and the cell cycle. These findings support existence of such interrelationship, although detailed mechanisms involved are not fully elucidated.
