Proteasome Inhibitors

Abstract:

Onemocnění různými typy rakoviny je v lidské populaci stále četnější. S rostoucím procentem výskytu roste též rezistence na běžně používáná léčiva. Z toho důvodu jsou vyvíjeny a testovány nové látky, které by pomohly při léčbě tohoto onemocnění. Vysoce efektivní strategií je inhibice proteazomu, která se úspěšně používá od roku 2003, kdy bylo schváleno první uznané léčivo, bortezomib (I. generace). Velké procento pacientů bylo na léčbu bortezomibem rezistentních, což vedlo k vývoji inhibitorů proteazomu II. generace (carfilzomib, ixazomib, marizomib, oprozomib a disulfiram). Inhibitory I. a II. generace jsou zaměřeny na 20S podjednotku proteazomu. Vykazují dobrou léčebnou odpověď, nicméně se poměrně často objevuje relaps onemocnění. Z tohoto důvodu se při hledání nových typů léčiv studie zaměřily především na 19S podjednotku proteazomu. Tak vznikly další typy potenciálních léčiv, které různým způsobem ovlivňují ubikvitin-proteazomový systém (UPS). Mezi tyto látky patří RA190, RA183, RIP-1, ubistatin B, capzimin, KDT-11, pevonedistat, serdemetan, nutlin-3 a NSC-207895, jenž v současné době procházejí intenzivním testováním. Je třeba zjistit, proč dochází k rezistenci nádorových buněk na používané léčivo a eliminovat případné vedlejší účinky.

Abstract:

Disease from various types of cancer is becoming more common in the human population. As the percentage of occurance increases, so does resistance to commonly used drugs. For this reason, agents are being developed and tested to help with the treatment of this disease. A highly effective strategy is proteasome inhibition, which has been used successfully since 2003, when the first authorized drug, bortezomib (generation I), was approved. A large percentage of patients were resistant to bortezomib, leading to the development of II. generation proteasome inhibitors (carfilzomib, ixazomib, marizomib, oprozomib and disulfiram). Inhibitors of I. and II. generations were focused on the 20S proteasome subunit. They showed good results, however, relapse often occurred. For this reason, in the search for new types of drugs, studies have focused on the 19S proteasome subunit. So the new potential drugs that variously target the ubiquitin-proteasome system (UPS) such as RA190, RA183, RIP-1, ubistatin B, capzimin, KDT-11, pevonedistat, serdemetan, nutlin-3 and NSC-207895 were formed.These potential drugs are currently under intensive investigation. It is necessary to find out why tumor cells are resistant to the used drugs and to eliminate their side effects.
