Native and denaturing egg proteins electrophoresis.

Abstract:

Cílem bakalářské práce optimalizovat přípravu polyakrylamidového a agarózového gelu, seznámit se s kvalitativním a kvantitativním zastoupením proteinů obsažených ve vaječném bílku, dále ověřit použitelnost elektroforézy pro analýzu reálných vzorků vaječných proteinů a to jak za denaturujících, tak i za nedenaturujících podmínek. Nakonec srovnat náročnost provedení a analytický přínos obou typu elektroforéz. Teoretická část práce přehledně popisuje strukturu jednotlivých proteinů obsažených ve vaječném bílku a zahrnuje také základní principy elektroforézy. Praktická část práce je zaměřena na optimalizaci přípravy polyakrylamidového a agarózového gelu a vzorků proteinů vaječného bílku. Dále zahrnuje popis provedení elektroforézy za denaturujících i nedenaturujících podmínek na obou typech nosičů. Součástí praktické části práce je také vytvoření návodu pro elektroforézu proteinů ve vzorku slepičího bílku, který bude využíván v praktickém cvičení z biochemie. Na základě získaných elektroforeogramů lze říct, že nevýhodou agorózového gelu oproti polyakrylamidovému je jeho nestálost, tloušťka a také menší rozlišovací schopnost. Naopak výhodou je jeho snadná příprava a nulová toxicita. Z analytického hlediska nativní elektroforéza oproti elektroforéze za denaturujících podmínek poskytuje ne tak detailní výsledky, ale její výhodou je naopak snadnější a jednoznačnější vyhodnocení získaných výsledků.

Abstract:

The aim of the bachelor thesis is to optimize the preparation of polyacrylamide and agarose gel, to get acquainted with the qualitative and quantitative composition of proteins contained in egg white. Further, to verify the applicability of electrophoresis for analysis of real samples of egg proteins under both denaturing and non-denaturing conditions. Finally, compare the performance and analytical contribution of both types of electrophoresis. The theoretical part of the work clearly describes the structure of individual proteins contained in egg white and also includes the basic principles of electrophoresis. The practical part of the work is focused on optimizing the preparation of polyacrylamide and agarose gel and egg white protein samples. It also includes description of electrophoresis under both denaturing and non-denaturing conditions on both types of carriers. Part of the practical part of the work is also the creation of instructions for electrophoresis of proteins in a sample of chicken protein, which will be used in practical exercises in biochemistry. Based on the obtained electrophoreograms, it can be said that the disadvantage of agorose gel compared to polyacrylamide gel is its instability, thickness and less resolution. On the contrary, the advantage is its easy preparation and zero toxicity. From the analytical point of view, native electrophoresis compared to electrophoresis under denaturing conditions provides not so detailed results, but its advantage is, on the contrary, easier and clearer evaluation of the obtained results.
