The determination of riboflavin in cereal products

Abstract:

V teoretické části diplomové práce je popsán vitamin B2 {--} riboflavin, jeho zdroje a vlast-nosti (fyzikálně-chemické i fyziologické) a také ztráty vitaminu při technologických opera-cích. Dále jsou popsány analytické metody, především chromatografické techniky, využí-vané pro stanovení tohoto vitaminu a technika HPLC. Je zde uvedena i charakteristika ce-reálních produktů. Experimentální část popisuje optimalizaci izolačního postupu vitaminu B2 z vybraných 22 běžných cereálních produktů (mouka, chléb, rohlík, cereální snídaně, těstoviny), kdy bylo zjišťováno vhodné složení a množství extrakčních činidel i průběh celé izolace. Následně byly pomocí chromatografické techniky HPLC/UV-DAD stanoveny obsahy riboflavinu ve vybraných 22 cereálních produktech, kdy obsah vitaminu klesal v následujícím pořadí: cereální snídaně, mouky obohacené vitaminy, celozrnné sušenky, celozrnné chleby, celo-zrnné mouky, pšeničný chléb, celozrnné těstoviny, tukový rohlík, pšeničné mouky a celo-zrnné pečivo.

Abstract:

Theoretical part of the thesis describes vitamin B2 {--} riboflavin, its sources and features (physico-chemical and physiological) and also losses of vitamin at technological operati-ons. Further there are described analytical methods, first of all chromatographic technics, to use for determination of vitamin B2 and HPLC technic. The characteristics of cereal pro-ducts also introduced. Experimental part describes optimalization of isolating process of vitamin B2 of 22 selec-ted common cereal products (flour, bread, roll, cereal breakfast, pastries), the suitable com-position and quantity of extraction reagents and isolation process were evaluated. Sub-sequently the contents of riboflavin of 22 selected cereal products with chromatographic technic HPLC/UV DAD was defined. The riboflavin content decreases is this order: cereal breakfast, flours enriched with riboflavin, wholemeal biscuits, wholemeal breads, whole-meals, bread, wholemeal pastries, roll, wheat flours and wholemeal rolls.
