Genetics of the Eurasian lynx of the Western Carpathians

Abstract:

Po rozsáhlém pronásledování v minulých staletí v Evropě se rys ostrovid (Lynx lynx) začíná pozvolna vracet do naší přírody. Na území České republiky, v Moravskoslezských Beskydech a Javorníkách, zasahuje okraj areálu rozšíření karpatské populace rysa ostrovida. Tato beskydsko-javornická populace je naší jedinou původní populací rysa a i když navazuje na hlavní zdrojovou populaci, její početnost je nízká a kolísá. Proto je důležité tuto populaci sledovat a zkoumat faktory, které ji nejvíce ohrožují, tedy pytláctví, migrační bariéry a fragmentaci krajiny. Cílem této studie bylo využít neninvazivní genetickou metodu pro zjištění početnosti, denzity a prostorové aktivity populace rysa ostrovida na vybraném území v České republice a na česko-slovenském pomezí – v Moravskoslezských Beskydech, Javorníkách, Hostýnsko-vsetínské hornatině a Vizovické vrchovině. Dalšími cíli bylo popsat genetickou variabilitu a strukturu studované populace a dozvědět se více o příbuznosti jedinců. Mezi roky 2009–2013 bylo ve sledovaném území nalezeno 156 vzorků rysího trusu, chlupů a moči. Analýza těchto vzorků na 19 mikrosatelitových lokusech umožnila identifikaci 18 jedinců rysa. Velikost celé populace byla odhadnuta na 10–15 jedinců, odhad efektivní velikosti populace byl 9,4 jedinců. Denzita byla stanovena na 0,66 jedince na 100 km2. Počet alel na jednotlivých lokusech se pohyboval mezi dvěma a šesti. Hodnoty průměrné pozorované a očekávané heterozygotnosti byly 0,631 a 0,583. Nebylo prokázáno vychýlení populace z Hardyho-Weinbergovy rovnováhy, populace se tedy jeví dostatečně velká, na to aby bylo možné pozorovat efekt inbreedingu. Faktorová korespondenční analýza však naznačila genetickou strukturovanost populace, kterou potvrdila i signifikantní hodnota indexu Fst (G-test, Fst= 0,096, P = 0,00167). Tuto strukturovanost pomohla objasnit analýza v programu ML-Relate a v Cervusu, kdy bylo zjištěno, že celá populace je tvořena ze dvou subpopulací, které se skládají ze dvou „rodinných“ skupin – javornické a beskydské.

Abstract:

After the long persecution during the last centuries in Europe, the Eurasian lynx (Lynx lynx) have begun to return slowly to our landscape. In the Czech Republic, in the Moravian-Silesian Beskids and Javorníky mountains, there is the edge of the species distribution in the Carpathians. This population of the Moravian-Silesian Beskids and the Javorníky mountains is the only one autochthonous population of lynx in the Czech Republic and although it is connected with the main source population in the Carpathians, its population size is low and fluctuating. Therefore, it is important to monitor the population and study factors which threaten it the most, i.e. poaching, migration barriers and habitat fragmentation. The main aim of this study was to estimate the population size, density and to analyse the spatial activity of the population of the Eurasian lynx in the study area using non-invasive genetic method in the Czech Republic and in the Czech-Slovakian borderland – in the Moravian-Silesian Beskids, the Javorníky mountains, the Hostýn-Vsetín Mountains and in the Vizovice Highlands. The next aim was to describe genetic variability and structure of the study population and kinship analysis. 156 samples of scat, hair and urine were collected in the study area during snow-tracking sessions between 2010–2013. Using in total 19 microsatellite loci and the sex specific marker SRY 18 individuals of lynx were identified. The total population size was estimated at 10–15 individuals, the effective population size at 9,4 individuals. Estimated density was 0,66 individuals per 100 km2. The number of alleles for each locus was between two and six alleles. The observed heterozygosity was Ho = 0,631 and the expected heterozygosity was He = 0,583. There was no significant deviation from Hardy-Weinberg expectations, therefore the population seems to be large enough for the possible observation of the inbreeding effect. Factorial correspondence analysis indicated that the population is genetical structured, what was confirmed also by significant value of Fst index (Fst = 0,096, P = 0,00167). The structure can be explained thanks to the kinship analysis in ML-Relate and Cervus, where was found that the population consists of two „family“ groups – the first one in the Javorníky mountains and the second one in the Moravian-Silesian Beskids.
