Cell motility in cryptosporidia (Apicomplexa)

Abstract:

Cílem předkládané bakalářské práce bylo pomocí kombinovaného přístupu světelné mikroskopie, fluorescenčního značení a biochemických postupů provést předběžnou analýzu cytoskeletálních útvarů a proteinů, které se podílí na pohybu sporozoitů jednobuněčného žaludečního parazita Cryptosporidium muris. První část této bakalářské práce se věnuje literární rešerši, která zahrnuje všeobecnou charakteristiku studovaného parazita a stručný rozbor buněčného pohybu výtrusovců se zaměřením na kryptosporidie. Jsou zde popsány organely, struktury a mechanizmy nezbytné pro jejich buněčný pohyb. Praktická část práce se zaměřuje na popis materiálu a použitých metodických postupů, výsledky s fotografickou dokumentací a diskuzi. Vzhledem k tomu, že jedním z cílů práce byla i standardizace protokolů použitých pro imunodetekci pohybových struktur u kryptosporidií, text také zahrnuje detailní postupy a návody na přípravu potřebných reagencií.

Abstract:

The main aim of submitted bachelor thesis was to perform a preliminary analysis of cytoskeletal elements and proteins that participate in cell motility of sporozoites of an unicellular gastric parasite Cryptosporidium muris using combined approach of light microscopy, fluorescence labelling and biochemical procedures. The first part of this thesis is dedicated to a literary research, which comprises general characteristics of studied parasite and a brief overview of cell motility of apicomplexans focusing on cryptosporidia. Organelles, structures and mechanisms essential for the cell motility are described herein. Practical part focuses on describing materials and used methods, results with photographic documentation and discussion. Considering that one of the set aims was the standardization of protocols used for immunodetection of motility structures in cryptosporidia, the text includes detailed description of procedures and instructions for preparation of required reagents.
