Analysis of processed cheese fortified with antioxidants

Abstract:

Tato práce se zabývá analýzou tavených sýrů obohacených přídavky antioxidantů - standardních látek rutinu, kvercetinu a rakytníkového oleje. Teoretická část práce zahrnuje charakteristiku tavených sýrů a rešeršní část, která popisuje aplikace různých látek (katechinů, silic, luteinu, karotenů, kyseliny askorbové) do mléčných výrobků. Dále jsou také uvedeny základní informace o rutinu, kvercetinu a rakytníku. V praktické části byl nejprve nalezen vhodný způsob zpracování vzorků před analýzou a také byly optimalizovány chromatografické podmínky pro analýzu rutinu a kvercetinu pomocí HPLC, které byly následně použity k identifikaci a kvantifikaci jednotlivých přídavků ve vzorcích tavených sýrů. Dále bylo provedeno stanovení antioxidační aktivity všech vzorků pomocí metod ABTS, DPPH a FRAP.

Abstract:

This thesis deals with an analysis of processed cheese enriched with the addition of antioxidants - standard compounds rutin, quercetin and sea buckthorn oil. The theoretical part includes characteristics of processed cheese and in research part, the application of various substances (catechins, essential oil, lutein, carotene, ascorbic acid) into dairy products are described. Furthermore, basic informations about rutin, quercetin and sea buckthorn are also given. In the practical part, at first a suitable method of processing the sample, and secondly the chromatographic conditions for analysis of rutin and quercetin by HPLC were found. These conditions were then used for identification and quantification the addition of antioxidants in samples of processed cheeses. Another part was determination of antioxidant activity of all samples using methods ABTS, DPPH and FRAP.
