Dependance of methylene blue aggregation extent on salt concentration in frozen aqueous solutions

Abstract:

Tato diplomová práce se zabývá studiem agregace methylenové modři v závis-losti na koncentraci anorganických solí (NaCl, CaCl2 a MgCl2) a cukrů (sacharóza a glukóza) v roztocích a po jejich zamrazení. Teoretická část práce je rozdělena do tří hlavních celků a jejím cílem je uvedení čtenáře do širšího kontextu prezentované problematiky. První celek obsahuje obecné informace o mrazení, především o vzniku mrazem zakoncentrovaného roztoku, ve kterém se výlučně odehrává veškerá kryochemie. Druhý celek se soustředí na možnosti stabilizace farmaceuticky aktivních látek, především lyofilizaci a vitrifikaci. Závěr teoretické části rozebírá agregaci molekul a možnosti jejího pozorování za použití methylenové modři. Experimentální část této práce je rozdělena na tři oddíly. První tvoří metodická sekce, která obsahuje výčet použitých chemikálií, popisuje přístroje a přípravu vzorků používaných v této práci. Druhý oddíl shrnuje výsledky na základě absorpčních spekter methylenové modři v roztoku a po zamrazení v závislosti na koncentraci solí a cukrů. Tyto výsledky tvoří základ pro závěrečnou diskusi. Výsledky této práce přináší nový vhled do studia agregace v roztoku a po za-mrazení. Agregace je jedním z jevů, díky kterému může docházet k degradaci farmaceuticky a biologicky aktivních látek. Tato práce přináší metodu, díky které je možné studovat agregaci způsobenou excipienty na poměrně levném systému bez nutnosti použití drahých aktivních látek. Zároveň tato práce zobecňuje, jak koncentrace excipientů ovlivní agregaci způsobenou mrazením.

Abstract:

This thesis deals with the study of the aggregation of methylene blue as a function of the concentration of inorganic salts (NaCl, CaCl2 and MgCl2) and sugars (saccharose and glucose) in a liquid solution and in a frozen state. The theoretical part of the thesis is divided into three main parts and aims to introduce the reader to the broader context of the presented problem. The first part contains general infor-mation about freezing, in particular it focuses on the formation of a freeze-concentrated solution, which constitutes the sole cryochemical reaction medium. The second part focuses on the possibilities of stabilization of pharmaceutically active substances especially via lyophilization and vitrification. Finally, the concept of aggregation and the possibilities of its observation utilizing methylene blue con-cludes the theoretical part. The experimental part of this thesis is divided into three sections. The first is the methodological section, which lists all the used chemicals, and describes the instru-ments, and sample preparation methods used in the course of this work. The second section presents results, i.e., the absorption spectra of methylene blue in solution and after freezing as a function of salt and sugar concentration. These results are further discussed in the last section of the experimental part. The results of this work provide a novel insight into the study of aggregation in solution and during freezing. Aggregation is one of the phenomena through which the degradation of pharmaceutically and biologically active substances can occur. This work presents a method that allows studying aggregation caused by excipients on a relatively inexpensive system without the need for expensive pharmaceutically active compounds. Furthermore, this work generalizes the concentration depend-ence of excipients on the final freezing-induced aggregation.
