An Influence of the Mutant Genotype for Methylentetrahydrofolat reductase on the Level of Homocysteine in the Patiens with Tromboembolism Affection. An Occurrence of 4G/5G Polymorphism in the Gene for Inhibitor of Activator Plazminogene 1 in the Region Ostrava.

Abstract:

Diplomová práce pojednává o vztahu mezi jednotlivými mutacemi v MTHFR genu a hladinou plazmatického homocysteinu. Zvýšená hladina homocysteinu ? homocysteinémie je jedním z rizikových faktorů vzniku tromboembolické nemoci a může být způsobena právě mutacemi v genu pro MTHFR. Druhá část práce je zaměřena na výskyt polymorfismu v genu pro PAI-1, o jehož přímém pozitivním vlivu na tromboembolickou nemoc se stále diskutuje. V teoretická části je probrána problematika pojmu mutace, mechanismy vzniku mutací, mechanismy vzniku arteriální i žilní trombózy a hlavní příčiny trombóz. Praktická část se zabývá jednotlivými typy vyšetřovacích metod mutací v genu pro MTHFR a polymorfismu PAI-1. Dále se zaměřuje na statistické zpracování výsledků vyšetření pacientů s tromboembolickými potížemi provedených v Krevním centru FN Ostrava od května 2006 do dubna 2007.

Abstract:

This diploma thesis deals with relationship between mutation in MTHFR gene and level of plasmatic homocysteine. Increased level of homocysteine - hyperhomocystinemia is one of the hazard factor creation thromboembolic diseases, which can be caused just mutation in MTHFR gene. Second part of this thesis is focused on incidence polymorphism in PAI-1 gene and about his possible positive influence on thromboembolic diseases. In theoretic part are discussed points regarding, mutation, mechanism of genesis mutations, and mechanism of genesis arterial and venous thrombosis and main causes of thrombosis. Practical part deals with particular processes investigative methods mutation in MTHFR gene and polymorphism PAI-1. Next focus is on statistical research mutation of patient of Blood centre Ostrava from May 2006 till April 2007.
