Telomeric chromatin in mutant Arabidopsis thaliana plants with defects in the histone methylations loading

Abstract:

Epigenetic modifications of DNA and histones play an important role in the regulation of gene expression and other key processes in the cell. They can also affect telomeres, the nucleoprotein structures protecting the ends of linear chromosomes. In theoretical part of this thesis, epigenetics of telomeres, especially modifications of telomeric histones are described. The experimental part is devoted to the study of Arabidopsis thaliana plants and their mutants suvh4 and ibm1 having T-DNA insertion in the appropriate gene. SUVH4 is a histone methyltransferase, which adds methyl groups to lysine 9 of histone 3 and IBM1 is H3K9 demethylase; in previous studies H3K9me2 was identified as dominant mark of plant telomeric histones. The impact of the loss of function of these enzymes on the ability to maintain a stable telomere length and telomerase activity was analyzed in three subsequent generations of plants. In the third generation of mutants, levels of specific epigenetic modifications on telomere histones were analysed. It was found out that in ibm1 mutants, telomeres were shortened, telomerase activity slightly decreased, but the level of transcription of the TERT gene remained unchanged. On the other hand, suvh4 mutants had reduced telomerase activity, which was correlated with reduced levels of TERT transcription, but without changes in telomere length. Lower levels ofH3K9me2 modification at telomeric histones were observed in both types of mutants.

Abstract:

Epigenetické modifikace DNA a histonů mají důležitou roli v regulaci genové exprese a jiných klíčových procesů v buňce. Mohou ovlivňovat i telomery, nukleoproteinové struktury chránící konce lineárních chromosomů. Tato diplomová práce se v teoretické části zabývá epigenetikou telomer, především modifikacemi histonů s nimi asociovanými. Experimentální část se věnuje studiu rostlin Arabidopsis thaliana a jejich mutantů suvh4 a ibm1, které mají v příslušném genu T-DNA inzerci. SUVH4 je histon methyltransferáza, která přidává metylovou skupinu na lysin 9 histonu 3 a IBM1 je H3K9 demetyláza; H3K9me2 byla v předchozích studiích identifikována jako dominantní modifikace telomerových histonů rostlin. Byl analyzován dopad ztráty funkce těchto enzymů na schopnost udržování stabilní délky telomer a aktivitu telomerázy ve třech následujících generacích rostlin. Ve třetí generaci mutantů byly sledovány i změny v hladinách konkrétních epigenetickýchmodifikací telomerových histonů. Bylo zjištěno, že u mutantů ibm1 byly telomery zkráceny, aktivita telomerázy mírně snížena, ale úroveň transkripce genu TERT se nezměnila. U mutantů suvh4 snížená aktivita telomerázy naopak korelovala se sníženou úrovní transkripce TERT, avšak bez změny délky telomer. V obou typech mutantů bylo pozorováno snížení hladiny modifikace H3K9me2 na telomerových histonech.
