In vitro models in testicticular toxicity assessment

Abstract:

V diplomové práci je nastíněna problematika narušení samčí reprodukce, která může být způsobená mnoha vnějšími vlivy včetně kontaktu člověka s chemickými látkami působícími nepříznivě na testikulární systém (varlata), což může vést až k neplodnosti. Teoretická část se zabývá využitím 3D in vitro modelů v hodnocení testikulární toxicity a v objasnění buněčných a molekulárních mechanismů vedoucích k narušení samčí reprodukce. Celá práce se zaměřuje zejména na jeden typ testikulárních buněk, na Leydigovy buňky, které hrají nezastupitelnou roli v produkci samčích pohlavních hormonů nezbytných pro správný vývoj varlat a produkci funkčních spermií. Shrnuje jejich vývoj a funkce ve zdravém organismu, dostupné zdroje těchto buněk pro in vitro modely spolu s jejich výhodami i nevýhodami, dále přináší informace o výhodách a nevýhodách dostupných 3D in vitro modelů a o jejich praktickém využití při hodnocení testikulární toxicity v toxikologii. Praktická část této práce je zaměřena na pokročilý 3D in vitro model multicelulárních sféroidů tvořený Leydigovými TM3 buňkami a na jeho využití při stanovení testikulární toxicity vybraných environmentálně relevantních kontaminantů životního prostředí (herbicid methoxychlor, produkty osobní potřeby triklosan a triklokarban), které jsou podezřelé z nepříznivého ovlivňování reprodukčního a endokrinního systému samců. Vliv látek byl hodnocen pomocí parametrů charakterizujících velikost, tvar a životaschopnost sféroidů. Triklosan neměl podstatný vliv na 3D model Leydigových buněk na rozdíl od 2D modelu. Naproti tomu methoxychlor sice nesnižoval životaschopnost buněk tvořících sféroid, ale způsoboval narušení jejich tvaru a kompaktnosti. Triklokarban vykazoval nejvyšší testikulární toxicitu a ovlivňoval nejen velikost a tvar sféroidů, ale i jejich životaschopnost.

Abstract:

The diploma thesis addresses the problem of the male reproduction which can be caused by many external factors including chemicals having adverse effects on testicular system (testis) which may lead to infertility. The theoretical part of this thesis deals with the use of the 3D in vitro models in the assasment of testicular toxicity and in the assessment of cellular and molecular mechanisms leading to impairment of male reproduction. The thesis is focused mainly on one type of testicular cells, on Leydig cells which play an important role in the production of male sex hormones essential for the development of testis and for the production of mature sperms. The thesis summarizes the development and functions of Leydig cells, the sources of this testicular cell type for in vitro models including their adventages and disadvantages and brings information on advantages and disadvantages of 3D in vitro models and on their usefulness for testicular toxicity assesment in toxicology. The practical part of the thesis is focused on an advanced 3D in vitro model of multicellular spheroids formed by Leydig TM3 cells and its application for the evaluation of testicular toxicity of environmentally relevant contaminants (herbicide methoxychlor, personal care products triclosan and triclocarban). The chemical impact was evaluated using parameters characterizeing size and shape and viability of spheroids. Triclosan had no significant impact on 3D model of Leydig cells, which is in contrast to 2D model. Methoxychlor did not decrease viability of cells forming spheroids but disturbed their shape and compactness. Triclocarban was the most potent testicular toxicants and affected not only the size and shape of spheroids, but also their viability.
