The investigation of chromosomal aberrations of miscarried foetuses by QF-PCR

Abstract:

Tato práce se zabývá vyšetřováním chromozomových aberací u potracených plodů pomocí metody QF-PCR. Konkrétně jsou vyšetřovány numerické aberace chromozomů 13, 15, 16, 18, 21, 22, X a Y. Práce je rozdělena na část teoretickou a část praktickou. V teoretické části jsou popsány metodiky prenatální diagnostiky, které jsou schopny s určitou mírou pravděpodobnosti odhalit některé numerické aberace. Po ní následuje kapitola týkající se potratů a jejich příčin Další část je věnována vzniku numerických aberací a klinickým projevům těchto aberací u živě narozených dětí. V závěru se nacházejí kapitoly popisující principy PCR a sekvenování. Obsahem praktické části je provedení QF-PCR a to včetně doporučení pro její správné provedení a vyhodnocování patologických a fyziologických výsledků. Dále je praktická část doplněna o statistickou kapitolu, v níž jsou zhodnoceny výsledky, které byly touto metodou zpracovány na Oddělení lékařské genetiky ve FN Brno v letech 2010 až 2012.

Abstract:

This thesis deals with the investigation of chromosomal aberrations of miscarried foetuses done by the QF-PCR method. In particular it examines the numerical aberrations of chromosomes 13, 15, 16, 18, 21, 22, X, and Y. The thesis is divided into two parts – theoretical and practical. In the theoretical part, the methods of prenatal diagnosis which with certain probability can detect some of the numerical aberrations are being described. This is followed by a chapter concerning miscarriages and their causes. The next part is then devoted to the formation of numerical aberrations and the clinical signs of these aberrations on children born alive. In the end the chapters describing the principles of PCR and sequencing are to be found. The content of the practical part is the application of the QF-PCR, including recommendations for its correct application and the process of evaluation of the pathological and physiological results. Furthermore, the practical part is supplemented by a chapter of statistics where the results which were achieved through this method at the Department of Medical Genetics of the University Hospital Brno between the years 2010 and 2012 are evaluated.
