Sample preparation for protein ubiquitination analysis

Abstract:

This thesis deals with an important post-translational modification, ubiquitination. An important part of ubiquitin, regulatory protein, is the N-terminus and seven lysine residues, through which originates mutual binding of ubiquitin molecules in the polyubiquitin chain. The various possibilities of binding provide diversity in the creation of polyubiquitin chains, and thus their significance. Since ubiquitinated proteins in the analyzed protein mixtures exist mostly as a minor component, the first important and difficult step is their correct separation and identification. Experimental part of this work deals with precisely this part of the research and thus contains optimization methods for isolating and identifying of ubiquitin proteins by antibodies on the peptide and protein level. It has been shown that the optimization takes more time and work, thus future progressin this area is expected.

Abstract:

Táto bakalárska práca sa zaoberá významnou posttranslačnou modifikáciou, ubiquitináciou. Významnými súčasťami ubiquitínu, regulačného proteínu, je N-koniec a sedem lyzínových zvyškov, cez ktoré dochádza k vzájomnému naväzovaniu molekúl ubiquitínu v polyubiquitínových reťazcoch. Rôzne možnosti naväzovania dodávajú rozmanitosť v tvorbe polyubiquitínových reťazcov, a tým ich významu. Keďže ubiquitinované proteíny sa v analyzovaných zmesiach proteínov väčšinou vyskytujú ako minoritná zložka, tak prvým dôležitým a zložitým krokom pri ich skúmaní je správna separácia a identifikácia. Experimentálna časť tejto práce sa zaoberá práve touto časťou výskumu a bola v nej prevedená optimalizácia metódy izolácie a identifikácie ubiquitinovaných proteínov pomocou protilátok na peptidovej a proteínovej úrovni. Ukázalo sa, že optimalizácia vyžaduje viac práce a času, preto sa v nej bude v blízkej dobe pokračovať.
