Role of primary cilia during development of craniofacial structures

Abstract:

Primární cilie, jakožto drobné nepohyblivé struktury vyrůstající na povrchu buněk, byly dlouho přehlížené, jako organely bez důležité funkce. V posledních letech se ovšem dostávají do popředí zájmu vědců jako součást většiny buněk a vykazující řadu specifických funkcí. Struktura primární cilie je velice podobná pohyblivým ciliím. Zahrnuje mikrotubulární axonemu, bazální tělísko a plazmatickou membránu s řadou receptorů. Jako neustále se obnovující a zanikající organela využívá k procesu svého sestavení intraflagelární transport. Je také místem interakce některých signálních drah, které hrají důležitou roli při embryonálním vývoji organismů. Jakoukoliv změnou morfologie či funkce primární cilie tak vznikají vývojové poruchy, které se souhrnně nazývají ciliopatie. Řada z těchto vývojových defektů se projevuje kraniofaciáním fenotypem. Cílem této bakalářské práce je sumarizace ciliopatií s kraniofaciálním defektem z pohledu defektů v primárních ciliích.

Abstract:

The primary cilia as a small immobile structures located on the surface of cells were for a long time aside of scientific attention as structures without any important function. Recently, however, they present actual topic of developmental biology because of their presence on surface of almost all cells and with numerous functions. The entire structure is very similar to motile cilia. It includes a microtubule axoneme, a basal body and plasma membrane with a number of receptors. As a constantly evolving and dissapearing organelle, it is using an intraflagellar transport for assembling itself. It is also involved in tuning of some signaling pathways that play an important role in the embryonic development of organisms. Any change or damage in primary cilium morphology or function results in a variety of diseases that are called ciliopathies. Many of these diseases are manifested by a craniofacial phenotype. The aim of this bachelor thesis is to summarize ciliopathies with altered craniofacial development in point of view focused on primary cilia.
