RAD sequencing methods in population genetics and evolution

Abstract:

Práce stručně shrnuje vývoj genetiky, jakožto nástroje v biologických studiích, a její metody potřebné v populační genetice a evoluci se zaměřením na přístup zvaný RADSeq (Restriction-site Associated DNA Sequencing). Práce se nejprve zabývá molekulárními základy dědičnosti, izolací DNA a jejich fragmentů a historickými milníky genetiky. Pozornost je dále věnována metodám studujícím příbuznost mezi organismy pomocí morfologických znaků a makromolekulárních markerů a v závěru jsou stručně popsány metody NGS (Next Generation Sequencing). Stěžejní a závěrečnou kapitolu tvoří jedna z nejnovějších metod sekvenování a to metoda RADSeq, což je sekvenování oblastí genomu vybraných na základě délky po enzymatickém restrikčním štěpení DNA. V práci popisuji několik modifikací metody RADSeq a stručný přehled aplikace přístupu ve vybraných vědeckých studiích zaměřených na obratlovce. Tento souhrn slouží zejména jako podklad pro nastudování základů této problematiky a pro tvorbu diplomové práce.

Abstract:

This Thesis briefly summarizes the development of genetics as a tool in biological studies. A history of genetic methods used in population genetics and evolution in the past is also introduced, as well as a focus on a modern RADSeq (Restriction-site Associated DNA Sequencing) methodology. The first part of the Thesis is focused on historical milestones of genetics including a work with DNA and molecular basics of heredity. This work also deals with methods using morphological features and macromolecular markers to estimate relationships between organisms. In addition to this, an array of modern NGS (Next Generation Sequencing) methods is briefly described. A key chapter of the Thesis dedicated to the RADSeq methodology, which includes a sequencing of genome regions, selected by their length after an enzyme cleavage of DNA. Several modifications of RADSeq methodology is discussed including an application example in vertebrates studies. This summary serves mainly as a general overview for those interested in a history of a work with genotypic and phenotypic information in biological studies and as a pre-step for my Diploma Thesis.
