Methods of liquid culture of plant explants

Abstract:

Kultivace rostlinných explantátů a jejich mikropropagace je využívána nejen při základním fyziologickém výzkumu, ale také při produkci sekundárních metabolitů nebo velkokapacitní produkci sazenic, a to jak u hospodářky významných polních plodin a lesních dřevin, tak i okrasných rostlin, ovocných stromů a keřů. Kultivace explantátů v tekutých médiích je dalším krokem ve zlepšování kvality i kvantity těchto technik a použití bioreaktorů je jeho logickým vyvrcholením. Kultivace v bioreaktorech umožňuje kultivaci za řízených podmínek, což však sebou přináší řadu nečekaných problémů a vyžaduje detailní porozumění rostlinné fyziologii, interakcím mezi vnějším prostředím a rostlinou a také vzájemným interakcím fyzikálních a chemických faktorů. Práce je zaměřena na popis základních principů kultivace explantátů v podmínkách in vitro, dále na faktory vnějšího prostředí ovlivňující růst explantátů, výčet typů bioreaktorů využitelných při kultivaci rostlinných explantátů a popis techniky měření indukované fluorescence chlorofylu. Součástí práce je také kultivační experiment, ve kterém byl použit bioreaktor RITA® pracující na principu dočasného zaplavování kultur a souběžného měření kvantového výtěžku fluorescence chlorofylu fluorimetrem MONI PAM 2000, které bylo použito jako indikátor fyziologického stavu explantátů.

Abstract:

Cultivation of explants and their micropropagation are used not only in basic physiological research, but also in the production of secondary metabolites or production of seedlings both in economically-important field crops and forest trees, ornamental plants, fruit trees, and bushes. Cultivation of explants in liquid media is the next step in the improvment of the quality and quantity of these techniques and the use of bioreactors is a logical culmination of such trend. Cultivation in bioreactors allows explant cultivation under controlled conditions, but this brings a lot of unexpected problems and requires a better understanding of plant physiology, the interaction between the external environment and plants and also the mutual interaction of physical and chemical factors. The work is aimed at the description of the basic principles of cultivation of explants in vitro, as well as the environmental factors affecting growth of explants, lists the types of bioreactors that might be available for crop cultivation, explants and description of the technique of induced chlorophyll fluorescence measurements. Part of the Bc. thesis describes an experiment in which RITA ® bioreactors operating on the principle of temporary flooding of cultures were used. Simultaneously, measurements of fluorescence quantum yield of photosystem II by fluorimeter PAM MONI 2000 were done. Quantum yield was used as an indicator of physiological status of explants.
