Abstract:

Zisky a ztráty genetického materiálu (dnes označovány jako CNV z anglického copy--¬number variation) jsou již dlouhou dobu spojeny s řadou syndromů, onemocnění a dalších patologických stavů. Pomocí celogenomových metod, jako je například aCGH, jsou postupně odhalovány další CNV, které mohou mít klinický význam. CNV nalezené pomocí aCGH by však měly být vždy potvrzeny nezávislou metodou a pro lepší objasnění klinického významu by měli být vyšetření podrobeni rovněž rodiče probanda. Tato diplomová práce se zabývá optimalizací metody relativní kvantifikace pomocí qPCR pro účely ověřování nálezů CNV, které byly detekovány metodou aCGH na Oddělení lékařské genetiky Fakultní nemocnice Brno. Metoda qPCR je srovnávána s metodou FISH, která je rovněž často používána pro validaci nálezů aCGH. Druhá část práce je zaměřena na použití qPCR v rutinní diagnostice pacientů s mentální retardací, poruchami autistického spektra, vrozenými vývojovými vadami a dalšími patologickými fenotypy.

Abstract:

Gains and losses of genetic material (commonly referred to as CNVs – copy-number variations) have for a long time been connected to a number of syndromes, disorders and other pathological states. Using whole-genome methods such as aCGH more CNVs, which can be clinically significant, are being revealed. However, CNVs detected by aCGH should always be validated by an independent method and both parents of the proband should be tested for the presence of CNV in order to clarify its clinical significance. This diploma thesis deals with the optimization of relative quantification by qPCR as a method for validating CNV calls detected by aCGH at The Department of Medical Genetics, The University Hospital Brno. qPCR is compared with FISH, another method commonly used to validate aCGH findings. The following part of the thesis is focused on the use of qPCR in routine diagnostics of patients with intellectual disability, autism spectrum disorders, congenital anomalies and other pathological phenotypes.
