Výskyt cytotoxických kmenů Escherichia coli u selat v závislosti na suplementaci diety

The occurrence of cytotoxic Escherichia coli strains in piglets as a function of dietary supplementation

Abstract:

Rozsáhlé a intenzivní používání doplňků v krmivech hospodářských zvířat ovlivňuje jejich střevní mikrobiom, což může mít negativní dopad jak na zdraví zvířat, tak i lidí. Tato bakalářská práce se zaměřuje na vliv různých zinkových suplementů přidaných do krmiva selat na zvýšený výskyt cytotoxických kmenů Escherichia coli. Práce zahrnuje experiment provedený na selatech ve věku 28 dnů, která byla rozdělena do skupin a podrobena suplementaci ZnO a nanočásticemi ZnA a ZnC. Vzorky výkalů byly sbírány a analyzovány s cílem identifikovat přítomnost cytotoxických kmenů E. coli. Analyzováno bylo 41 izolátů a byl sledován výskyt čtyř genů, a to konkrétně genu eae, genu cnf, genu cdt a genu pks. Všechny tyto geny jsou typické pro cytotoxické kmeny E. coli. K analýze byla použita metoda PCR s provedením následné gelové elektroforézy a detekcí pod ultrafialovým světlem. V této práci byl potvrzen mírný vliv suplementace krmiva na výskyt genu eae. Bylo zjištěno, že v den 0 byl gen eae pozitivně detekován u 20 % vzorků ze skupiny kontrolní i skupiny, která byla suplementována nanočásticovou formou ZnC. V případě skupin suplementovaných ZnO a nanočásticemi ZnA nedošlo v den 0 k žádné pozitivní detekci. Desátý den odběru vzorků byla frekvence genu eae potvrzena u 50 % vzorků odebraných ze skupiny selat suplementovaných ZnO, zatímco u skupiny suplementované nanočásticemi ZnA byla frekvence výskytu pouze 12,5 %. V kontrolní skupině a skupině suplementované nanočásticemi ZnC nebyl tento gen detekován. U ostatních sledovaných genů nedošlo k žádnému pozitivnímu nálezu. Výsledky této studie přispějí k lepšímu pochopení účinku suplementace zinkem, jako prostředku ke snížení infekcí střevního traktu u odstavených selat.

Abstract:

Extensive and intensive use of supplements in the feed of livestock affects their intestinal microbiome, which can have a negative impact on both animal and human health. This bachelor thesis focuses on the influence of various zinc supplements added to piglet feed on the increased occurrence of cytotoxic strains of Escherichia coli. The study involved piglets aged 28 days, divided into groups and subjected to supplementation with ZnO and zinc nanoparticles (ZnA and ZnC). Fecal samples were collected and analyzed to identify the presence of cytotoxic strains of E. coli. A total of 41 isolates were analyzed, with the occurrence of four genes specifically examined: the eae gene, cnf gene, cdt gene, and pks gene, all characteristic of cytotoxic strains of E. coli. PCR method with subsequent gel electrophoresis and detection under ultraviolet light was used for analysis. The study confirmed a slight influence of feed supplementation on the occurrence of the eae gene. It was found that on day 0, the eae gene was positively detected in 20 % of the samples from both the control group and the group that was supplemented with nanoparticle form of ZnC. In the case of the groups supplemented with ZnO and ZnA nanoparticles, there was no positive detection on day 0. On the tenth day of sampling, the frequency of the eae gene was confirmed in 50 % of the samples taken from the group of piglets supplemented with ZnO, while the frequency of occurrence was only 12.5 % in the group supplemented with ZnA nanoparticles. This gene was not detected in the control group and the group supplemented with ZnC nanoparticles. There were no positive findings for the other monitored genes. The results of this study will contribute to a better understanding of the effect of zinc supplementation as a means of reducing intestinal tract infections in weaned piglets.
