The role of phosphorylation and dephosforylation in regulation of telomerase activity

Abstract:

Enzyme telomerase is a subject of interest in many researches not only because of its role in solving the so-called end replication problem but also because of its close relationship with a number of cancerous diseases. Closer clarification of its regulation would greatly help the progress of modern medicine. This thesis maps knowledge about phosphorylation and dephosphorylation events influencing telomerase activity. T-DNA insert mutants in the proteinphosphatase 2A regulatory subunit gene, the promotor of RUNKEL kinase gene and parainal genes TRA1A and TRA1B were selected. The length of telomere of Arabidopsis thaliana mutant plants with T-DNA insertion in the TRA1A and TRA1B genes that code for the phosphatidylinositol kinase was analyzed. No significant effect on the telomere length of the phosphatidylinositol kinases tested was found in either the first or the second generation of mutant plants. The inclusion of T-DNA in the protein phosphotase 2A subunits appeared to be lethal, as all the plants grown from these mutant lines had a wild genotype.

Abstract:

Enzým telomeráza je predmetom záujmu mnohých výskumov nielen kvôli úlohe v riešení tzv. koncovo replikačného problému, ale aj kvôli jej úzkemu vzťahu s radom nádorových ochorení. Bližšie objasnenie jej regulácie by výrazne pomohlo pokroku modernej medicíny. V tejto bakalárskej práci boli zmapované poznatky o fosforylačných a defosforylačných dejoch ovplyvňujúcich aktivitu telomerázy. Boli selektovaní mutanti s T-DNA inzerciou v génoch pre regulačné podjednotku proteinfosfatázy 2A, v promotore génu pre kinázu RUNKEL a v paralógnych génoch TRA1A a TRA1B. Bola analyzovaná dĺžka telomér mutantných rastlín Arabidopsis thaliana s T-DNA inzerciou v génoch TRA1A a TRA 1B, ktoré kódujú fosfatidylinozitol kinázy. Žiadny výrazný vplyv na dĺžku telomér u testovaných fosfatidylinozitol kináz sa nezistil ani u prvej ani pri druhej generácie mutantných rastlín. Inzercia T-DNA v génoch pre podjednotky proteinfosfatázy 2A sa javila ako letálna, pretože všetky vypestované rastliny z týchto mutantných línií mali divoký genotyp.
