Inaktivace nádorového supresoru p53 je důležitým krokem v nádorové transformaci mnoha typů buněk. Nejčastěji ji bývá dosaženo mutacemi v genu pro p53, ale u mnoha typů nádorů je za inaktivaci p53 zodpovědná zvýšená exprese onkogenu Mdm2 nebo blízce příbuzného onkogenu MdmX, která vede k inhibici transkripční aktivity p53, případně k jeho degradaci prostřednictvím 26S proteazomu. Inactivation of the p53 tumour suppressor is an important step in the oncogenic transformation of many cell types and is most commonly achieved by mutations in the p53 gene.
