Abstract:

Řada prací ukazuje, že dochází ke zvýšenému výskytu poruch samčího reprodukčního systému. Díky komplexnosti tohoto systému může být jeho narušení zapříčiněno více faktory. Jedním z environmentálních faktorů, které k tomuto snížení mohou přispívat, jsou kontaminanty ovzduší. Jedním z možných mechanismů působení atmosférických polutantů je narušení mezibuněčné komunikace ve varlatech. Tato práce se zabývá studiem vlivu vybraných zástupců polyaromatických uhlovodíků, které se běžně vyskytují ve vzduchu, na mezibuněčnou komunikaci pomocí mezerových spojů u Leydigových buněk zodpovědných za produkci samčích steroidů. Pro studium komunikace byla využita nenádorová buněčná linie TM3 odvozená z myších Leydigových buněk. Mezibuněčná komunikace byla stanovena metodou sledování migrace fluorescenčního barviva mezerovitými spoji po naříznutí buněčné monovrstvy (scrape-loading/dye transfer). Výsledky ukazují, že některé polyaromáty (1- methylantracen, 9-methylantracen, fluoren, fluoranthen a benzo[a]pyren) významně inhibují GJIC zatímco jiné ne (anthracen, 2-methylantracen). Studie částečně potvrdila souvislost mezi strukturou PAH a jejich účinkem na hladinu GJIC. Atmosferické polyaromáty jsou tedy schopny ovlivnit mezibuněčnou komunikaci u Leydigivých buněk in vitro, což by mohlo vést k narušení produkce androgenů a následně samčí reprodukce.

Abstract:

Many studies indicate that there is a growing incidence of malfunctions of male reproductive system. Due to complexity of the system there are many potential factors that could play a role in the observed malfunctions. One of these factors is effect of atmospheric contaminants on intercellular communication in testes. This work is focused on study of effect of several polyaromatic hydrocarbons on gap junction intercellular communication (GJIC) among Leydig cells, which play a crucial role in production of male steroid hormones. We used cell line TM3, which is derived from immature mouse Leydig cells. Intercellular communication was assessed using scrape-loading/dye transfer method. Our results show that some polyaromates (1-methyl antracene, 9-methyl antracene, fluorene, fluoranthene a benzo[a]pyrene) significantly inhibit GJIC but others do not (anthracene, 2-methylantracene). The study partially confirmed link between structure of PAH and their effect on GJIC . Thus, atmospheric PAH were able to affect GJIC in Leydig cells in vitro and this might lead to impairment of androgen production and subsequently even male reproduction.
