Bioinformatical and functional characterisation of the ARCTICA gene in Arabidopsis thaliana

Abstract:

Jasně definovaná a někdy polárně orientovaná lokalizace proteinů na určitých buněčných místech je u rostlin častou podmínkou jejich funkce spřaženou s globálním programem ontogeneze. Účelem této studie je charakterizovat, s podporou bioinformatických nástrojů, rodinu proteinů, označovaných jako ARCTICA, které se polárně projevují na plazmatické membráně buněk Arabidopsis thaliana. Jsou představeny vzory exprese ARCTICA genů v některých orgánech a tkáních Arabidopsis, jakými jsou: děložními lístky, kořeny a průduchy. Rovněž je prokázána i vnější laterální polární lokalizace proteinů Arctica v epidermálních buňkách kořene. Naše experimenty potvrzují redukci velkosti a růstové defekty u double mutantů genů ARCTICA. Tyto rostliny rovněž vykazují vady ve vývojových procesech, jako je tvorba průduchů a kořenových vlásků nebo vývoj vaskulaturního systímu. Kromě toho tato práce obsahuje některé základní poznatky o mechanismu molekulárních složek, které by mohly přispět k vnější polární lokalizaci ARCTICA v buňkách pokožky kořene buněk. Je však zřejmé, že tato problematika ještě bude vyžadovat další zkoumání.

Abstract:

Clearly defined and sometimes polar localization of proteins at distinct cells sites is often a prerequisite of their function in conducting the overall ontogenetic program of the plant. The purpose of this study is to characterise, with support of bioinformatic tools, a family of proteins, designated ARCTICA, showing a polar enrichment at the plasma membrane of Arabidopsis thaliana cells. We present the expression patterns of ARCTICA genes in Arabidopsis organs and tissues such as: cotyledons, roots and stomata. We also show the outer lateral polar localization of Arctica proteins in root epidermal cells. Our experiments illustrate that ARCTICA double mutants have reduced size and growth defects. Those plants also show defects in developmental processes such as stomatal formation and root hair as well as vasculature development. In addition, this work provides some initiall insights on the mechanisms and molecular components that might contribute to the outer polar localization of ARCTICA in epidermis root cells. However, it is evident that this topic will require further analysis.
