Ekotoxicita komplexních vzorků vodních květů sinic

Abstract:

Působení cyanotoxinů, zejména microcystinů, na různé vodní organismy je intenzivně zkoumáno. Zároveň se však stále častěji objevuje myšlenka, že toxicita komplexní biomasy sinic není způsobena jen známými cyanotoxiny. Významný negativní vliv mohou mít takové jevy, jako nedostatek kyslíku a vznik amoniaku při rozpadu biomasy, ale zejména přítomnost dalších dosud neidentifikovaných toxinů nebo jiných složek buňky sinic s toxickým působením. V současné době probíhají experimenty s cílem blíže identifikovat tyto látky. Významný toxický potenciál mohou mít mj. lipopolysacharidy, jejichž hlavním mechanismem působení je ovlivnění detoxikace cyanotoxinů, a látky peptidické povahy, které především inhibují proteázy u zooplanktonu. V této práci byla testována toxicita sedmi různých vzorků komplexní biomasy sinic a jejich frakcí. Vzorky biomasy pocházely z různých vodních ploch, měly odlišné taxonomické složení i obsah microcystinů. Frakcí bylo připraveno od každého vzorku pět a to: homogenizovaná komplexní biomasa, pelet a supernatant, získané centrifugací, a permeát a eluát, získané ze supernatantu pomocí extrakce na pevnou fázi. Převažující metodou testování byly testy akutní toxicity na hrotnatkách druhu Daphnia magna. Testovány byly tyto koncentrace : 100, 200, 400, 800 a 1200 mg/l. Získané výsledky potvrdily předpoklad, že toxicita sinic není způsobena jen známými cyanotoxiny (microcystiny). Jako příklad lze uvést silné toxické působení (48h-EC50 komplexní biomasy = 112,5 mg/l) vzorku bez detekovaných microcystinů v kontrastu s nízkou toxicitou vzorku s relativně vysokým obsahem microcystinů (48h-EC25 komplexní biomasy = 667 mg/l). Dále byl proveden také test chronické toxicity na Daphnia magna, použity byly frakce jednoho ze vzorků. Koncentrační řadu tvořily koncentrace 5, 15, 45, 135 a 405 mg/l. V komplexní biomase a supernatantu byl pozorován pozitivní vliv na růst a porodnost, v eluátu byl růst snížen a také začátek reprodukce byl opožděn a množství potomků bylo nízké.

Abstract:

The impact of cyanotoxins, especially of microcystins, on various water organisms has been intensively studied. At the same time there has been appearing an idea that toxicity of the complex biomass is not caused only by generally known cyanotoxins. Other phenomena, such as lack of oxygen and rise of amoniac, developing during the decay of the biomass, but also occurence of other non-specified toxins or other parts of the cyanobacteria cell with toxic effect can play an important negative role. Experiments focusing on closer identification of these substances have been currently done. Among others, two groups of substances can have an important toxic potential: lipopolysacharids, whose main mechanism of effecting consists in influencing the detoxication of cyanotoxins, and substances of peptidic character that particularly inhibit proteases in zooplankton. In this thesis the toxicity of seven different samples of the complex biomass and its fractions was tested. The samples came from different water reservoirs, had different taxonomic composition and different content of microcystins. The folloving fractions, five of each, were prepared: homogenized complex biomass; pellet and aqueous extracts prepared by centrifugation; permeate and eluate gained from the supernatant by means of solid phase extraction. The prevailing method of testing were tests of acute toxicity on Daphnia magna. The concentrations 100, 200, 400, 800 and 1200 mg/L were tested. The results confirmed the premiss that the toxicity of cyanobacteria is not caused only by identified cyanotoxins. For example, strong toxic impact (48h-EC50 of the complex biomass = 112,5 mg/L) of the sample without any microcystins contrasted with low toxicity of the sample with relatively high content of microcystins (48h-EC25 of the complex biomass = 667 mg/l). Besides that a test of chronic toxicity on Daphnia magna, for which one fraction of one sample was used, was made too. The concentrations 5, 15, 45, 135 and 405 mg/L were tested. In the complex biomass and the supernatant a positive influence on the growth and the birth rate was noticed. In the eluate the growth fell down, the start of the reproduction was delayed and the number of descendants was low.
