Gene expression related to phenolic compounds biosynthesis

Abstract:

Teoretická část této bakalářské práce se zabývá fenolovými látkami (FL), biosyntézou FL a enzymy spojenými s biosyntézou FL. Práce shrnuje ochranné strategie rostlin vůči biotickému a abiotickému stresu, zejména stresu způsobeném vysokou ozářeností. S ochranou proti světelnému stresu souvisí i molekuly, které detekují světlo - tzv. fotoreceptory. Jejich hlavní skupiny, struktury a funkce jsou popsány v poslední kapitole teoretické části. V rámci experimentální části byla provedena analýza exprese genů souvisejících s biosyntézou FL metodou RT-qPCR. Pro vlastní sledování byly zvoleny tři cílové geny: dihydroflavonol 4-reduktáza (HvDFR), chalkon izomeráza (HvCHI) a ?avonoid 3´,5´-hydroxyláza (HvF3´5´H), jejichž finální produkty - enzymy mají v konečném důsledku značný vliv na akumulaci FL, které jsou známy jako látky s výraznými antioxidačními účinky. Cílem této práce bylo zjistit, do jaké míry je ovlivněna exprese cílových genů spektrální kvalitou dopadajícího záření, při kterém byly rostliny ječmene setého (Hordeum vulgare) pěstovány. Z výsledků RT-qPCR vyplývá, že výrazně zvýšená exprese cílových genů byla pozorována zejména u rostlin pěstovaných na modro-zeleném světle. Podobný, jen méně výrazný efekt byl pozorován i v případě kultivací na modro-červeném a modrém světle. Pro doplnění byl proveden další experiment, při kterém byly rostliny pěstovány pod modrým světlem při různé teplotě (12 °C, 22 °C a 32 °C). I na těchto vzorcích pak byla provedena RT-qPCR analýza. U všech 3 sledovaných genů byla pozorována nejvyšší exprese při 22 °C.

Abstract:

The theoretical part of this bachelor thesis deals with phenolic compounds (PCs), PCs biosynthesis, and enzymes associated with PCs biosynthesis. The thesis summarizes the defence strategies of plants against biotic and abiotic stress, especially the stress caused by high irradiation. The molecules that detect light - so called photoreceptors - are also related to protection against light stress. Their main groups, structures, and function were described in the last chapter of the theoretical part. In the experimental part, an analysis of the expression of genes related to PCs biosynthesis was performed by RT-qPCR. Three target genes were selected: dihydroflavonol 4-reductase (HvDFR), chalcone isomerase (HvCHI), and ?avonoid 3´,5´-hydroxylase (HvF3´5´H), whose final products - enzymes have a significant effect on the accumulation of PC, which are known to have significant antioxidant properties. The aim of this thesis was to determine the extent to which the expression of the target genes is affected by spectral quality of light in which barley plants (Hordeum vulgare) were grown. The results of RT-qPCR show that significantly increased expression of target genes was observed especially in plants grown in blue-green light. A similar, only less pronounced effect was observed in the case of cultivation on blue-red-, and blue light. In addition, another experiment was performed in which the plants were grown under blue light at different temperatures (12 °C, 22 °C, and 32 °C). The RT-qPCR analysis was also performed on these samples. The highest expression was observed for all 3 genes in all samples, that were cultivated at 22 °C.
