Development of Capillary Electrophoresis and MALDI-TOF Mass Spectrometry Methods

Abstract:

ABSTRAKT První část Disertace se zabývá analytikou extraktů a farmaceutických produktů různými technikami, především vývojem metod kapilární zónové elektroforézy (CZE) a MALDI TOF hmotnostní spektrometrie (hmotnostní spektrometrie s laserovou desorpcí a ionizací pomocí matrice a analyzátorem doby letu). Ačkoliv se pro analysy složek Gingko biloba a produktů obsahujících Huperzine A používají většinou metody HPLC, má jejich použití zejména pro analýzy těchto tradičních látek čínské medicíny řadu omezení a nevýhod, jako jsou: dlouhá doba analýzy, nedostatečné rozlišení a krátká životnost chromatografických kolon v důsledku jejich snadné kontaminace. Proto bylo studováno použití CZE, a to jak pro charakterisaci a separaci komponent Ginkgo biloba extraktů (farmaceutické produkty) tak také výrobků jakými jsou např. Huperzine A tabletky. Byly vyvinuty nové analytické postupy. Pro optimalisaci separačních podmínek byla aplikována technologie kombinace plánování pokusů (ED) a umělých neuronových sítí (ANN). Je mnoho typů plánů pokusů: CCD, faktorový plán, redukované faktorové plány, atd. V práci bylo ukázáno, že faktorový plan s centrálním bodem poskytuje dostatečná data pro ANN model a jeho použitím je tak značně redukován nutný počet měření pro optimalisaci. Tento postup byl vyvinut a validován pro analysu Ginkgo biloba extraktů, farmaceutických produktů a pro stanovení Huperzinu A v různých matricích. Druhá část disertace je věnována studiu možností MALDI-TOF MS pro analysu vybraných mykotoxinů v ječmeni, sladu a v potravinách. Tato technika umožňuje vysoce citlivé a rychlé stanovení a/nebo detekci detection při odběru velmi malého množství vzorku. Byly vyvinuty nové postupy pro detekci a identifikaci vybraných mykotoxinů. Vypracovaná metoda otevírá možnosti pro zpracování velkého množství vzorků a tím monitorování kvality ječmene, sladu a potravin.

Abstract:

ABSTRACT The first part of the Thesis deals with evaluation of extracts and pharmaceutical products by means of various analytical techniques and development of Capillary Zone Electrophoresis (CZE) and Matrix Assisted Laser Desorption/Ionisation Time of Flight mass spectrometry (MALDI-TOF MS) methods. Although HPLC is mostly used for analysing the constituents of Ginkgo biloba and Huperzine A products, the use of HPLC for the analysis of traditional Chinese medicines often has many shortcomings such as: long analysis time, low resolution and short column lifetime due to easy contamination. Therefore, CZE was studied to be used for the characterization and separation of Ginkgo biloba extract (pharmaceutical products) and Huperzine A tablet products. New analytical procedures were developed. For optimization of separation conditions, Experimental design (ED) combined with Artificial Neural Network (ANN) was used. There are several types of experimental design: central composite design, factorial design, fractional factorial designs, etc. Factorial design with a central point has been proved to produce sufficient input data for ANN approximation and thus reduced the number of experiments necessary for optimisation. This approach was developed and validated for the analysis of Ginkgo biloba extracts and pharmaceutical product determination of the Huperzine A in different media. The second part of the Thesis is devoted to study the possibilities of MALDI-TOF MS for analysis of selected mycotoxins in barley, malt and food products. This techniqe enables highly sensitive and fast analysis and/or detection using very small samples. New methods for detection and identification of selected mycotoxins were developed. This method opens up possibilities for high throughput analysis and quality monitoring of barley, malt and food.
