Determination of Selected Amino Acids and Fatty Acids in Breast Milk

Abstract:

Jelikož v posledních letech je snaha o minimalizaci nákladů na transport, manipulaci, ale hlavně analýzu vzorků, byla vyvinuta technika splňující tyto požadavky, kterou je technika suché kapky. Ta je známá především z novorozeneckého screeningu, nicméně ji lze aplikovat i na vzorky mateřského mléka. Tato práce se zabývá stanovením hladin mastných kyselin a aminokyselin ve vzorcích mateřského mléka a kolostra získaných technikou suché kapky. Hladiny byly porovnány s hladinami publikovanými v odborné literatuře. Mastné kyseliny byly převedeny na odpovídající methylestery a analyzovány metodou plynové chromatografie s plamenově-ionizační detekcí. Aminokyseliny byly derivatizovány s naftalen-2,4-dikarboxaldehydem za vzniku odpovídajících 1-kyano-2-substituovaných-benz[f]isoindolových derivátů, které byly analyzovány metodou vysokoúčinné kapalinové chromatografie s fluorescenční detekcí.

Abstract:

Since in recent years there has been an effort to minimize the costs of transport, handling, but mainly for the analysis of samples, a technique of dried spot has been developed. It meets these requirement. This method is known mainly for neonatal screening, however, it can also be applied to breast milk samples. This work deals with the determination of levels of fatty acids and amino acids in samples of breast milk and colostrum obtained by the technique of dried spot. The levels were compared with the levels published in the literature. The fatty acids were converted to the corresponding methyl esters and analysed by gas chromatography with flame ionization detection. Amino acids were derivatized with naphthalene 2,3 dicarboxaldehyde to give the corresponding 1-cyano-2-substituted-benz[f]isoindole derivatives, which were analysed by high performance liquid chromatography with fluorescence detection
