Analytical characteristics of detection of oligoclonal IgG bands in cerebrospinal fluid and serum

Abstract:

Bakalářská práce se věnuje problematice oligoklonálních IgG pásů v likvoru a séru. V teoretické části se zaměřuje na obecnou likvorologii, intrathekální odpověď a její detekci, kvantitativní stanovení intrathekální odpovědi a metodu izoelektrické fokusace. V praktické části byla posouzena shoda mezi hodnotícími a mezi dvěma metodami detekce (imunofixace a afinitní imunoblotting). Dále byla posouzena reprodukovatelnost metody isoelektrické fokusace. Cílem práce je zjistit, jaká je shoda mezi hodnotícími, a tím určit obtížnost v interpretaci výsledků získaných touto metodou. Dalším cílem je porovnat mezi sebou dvě odlišné metody detekce oligoklonálních IgG pásů. Posledním cílem je posouzení reprodukovatelnosti metody izoelektrické fokusace.

Abstract:

The bachelor thesis deals with oligoclonal IgG bands in CSF and serum. In the theoretical part, it focuses on general CSF analysis, intrathecal response and its detection, quantitative determination of the intrathecal response and method of isoelectric focusing. In the practical part, the interobserver agreement and agreement between two methods for detection (immunofixation and affinity immunoblotting) was evaluated. Furthermore, the reproducibility of the isoelectric focusing method was assessed. The aim of the thesis is to find out the interobserver agreement and thus determine the difficulty in interpreting the results obtained by this method. Another objective is to compare two different methods for detecting oligoclonal IgG bands. The last objective is to assess the reproducibility of the isoelectric focusing method.
