Study of Metallome in Selected Cancer Diseases

Abstract:

Disertace je zaměřena na studium účinků kovů na nádorovou tkáň. Již na příkladu vztahu zinku a prostaty bylo objasněno, že hladiny kovů mohou významně ovlivňovat chování nádorových buněk. V tomto případě však dosud nebylo uspokojivě vysvětleno, zda role zinku je spíše tumor inhibující, nebo tumor protektivní. Studie byla prováděna na tkáňových kulturách a nově vytvořeném modelu zinek rezistentních linií odvozených od karcinomu prostaty (linie PNT1A, 22Rv1 a PC-3). Pomocí mikroskopických, molekulárně biologických a dalších biochemických metod byly popsány vlastnosti buněk rezistentních k zinečnatým iontům. Byly stanoveny faktory významné pro proces rezistence, jako jsou apoptóza, autofagie nebo metabolismus kovů, a také vznik pluripotence a profily genové exprese související s rezistencí. V práci byly popsány také biomechanické vlastnosti, metabolismus aminokyselin a pozorován byl i metastatický potenciál zinek rezistentních buněčných linií. Vytvořené zinek rezistentní linie vykazovaly agresivnější charakter než jejich protějšky, které nebyly vystaveny působení zinečnatých iontů. Dlouhodobá rezistence vzniká díky odlišným mechanismům, než jsou ty využívané při krátkodobém působení a jejich zapojení vede ke změnám ve fenotypu prostatických buněk. Dlouhodobé působení zinečnatých iontů má také vliv na změny v aminokyselinovém profilu a zvyšuje metastatický potenciál buněk. Na základě získaných dat bylo možné lépe pochopit vliv kovů na nádorovou buňku a diskutovat případné využití zinku jako terapeutického nástroje při prevenci a léčbě karcinomu prostaty.

Abstract:

This dissertation thesis is focused on the study of effects of metals on tumour tissue. It has already been elucidated for example by the relationship of zinc and prostate that the levels of metals can significantly influence the behaviour of tumour cells. In this case, however, it has not yet been satisfactorily explained whether the role of zinc is more tumour inhibiting or tumour protective. The study was performed on standard tissue cultures and a newly created model of zinc resistant cell lines derived from prostate cancer (PNT1A, 22Rv1 and PC-3 cell lines). Microscopic, molecular biological and other biochemical methods have been used to describe the properties of the cells resistant to zinc ions. Factors important for rezistance such as apoptosis, autophagy or metal metabolism, as well as the development of pluripotency, were determined, as well as expression profiles of zinc-rezistant cells. Biomechanical properties, amino acid metabolism and metastatic potential of zinc-resistant cell lines were also described. The created zinc-resistant cell lines were more aggressive than their counterparts that were not exposed to zinc ions. The long-term resistance results from different mechanisms than those used in short-term exposure and their involvement leads to changes in the prostatic cells phenotype. Prolonged action of zinc ions also affects changes in amino acid profile and increases the metastatic potential of cells. Based on the obtained data, it was possible to better understand the influence of metals on tumour cells and discuss the possible use of zinc as a therapeutic tool in the prevention and treatment of prostate cancer.
