Interactions of 5-hydroxytryptamine with myeloperoxidase and their role in modulation of neutrophil activity

Abstract:

Tato diplomová práce se zabývá studiem interakcí 5-hydroxytryptaminu s myeloperoxidasou a možnou modulací aktivity neutrofilů. Myeloperoxidasa je velmi účinným enzymem primárních granulí neutrofilů, účastní se jejich oxidativního vzplanutí a je schopna oxidovat biomolekuly s/bez zachycení na neutrofilních extracelulárních pastech. V místě zánětu se kromě neutrofilů vyskytuje také 5-hydroxytryptamin uvolňovaný z aktivovaných krevních destiček. Důležitým faktorem ovlivňujícím aktivitu myeloperoxidasy je pH mikroprostředí, které mění její katalytický cyklus, a tím i interakci s 5-hydroxytryptaminem. Při fagosomálním pH 5-hydroxytryptamin snižoval chemiluminiscenci a choval se tedy jako antioxidant. Naopak za fyziologického pH byla chemiluminiscence zvýšena v závislosti na koncentraci 5-hydroxytryptaminu, který se choval v tomto případě jako substrát myeloperoxidasy. Podobný efekt byl pozorován u N-acetyl-5-hydroxytryptaminu. V interakci s izolovanými neutrofily bylo zjištěno snížení chemiluminiscence u nejvyšší použité koncentrace 5-hydroxytryptaminu (10 µM) poukazující na jeho antioxidační vlastnosti. V navazujících experimentech se podařilo indukovat tvorbu neutrofilních extracelulárních pastí a vizualizovat strukturu DNA spolu s myeloperoxidasou a elastasou pomocí konfokálního mikroskopu. Ke zjištění, zda 5-hydroxytryptamin ovlivňuje tvorbu extracelulárních pastí, byla použita metoda oxidace 3,3´,5,5´-tetramethylbenzidinu pomocí myeloperoxidasy. Tato metoda naznačila snížení tvorby struktur v přítomnosti 5-hydroxytryptaminu, avšak tento poznatek vyžaduje potvrzení dalšími experimenty. Tato práce dokazuje závislost interakce 5-hydroxytryptaminu s myeloperoxidasou na pH. 5-hydroxytryptamin se v přítomnosti myeloperoxidasy choval jako antioxidant (při fagosomálním pH) a jako alternativní substrát (za fyziologického pH) a může tedy modulovat aktivitu neutrofilů prostřednictvím myeloperoxidasy.

Abstract:

This thesis deals with the study of interactions of 5-hydroxytryptamine with myeloperoxidase and a possible neutrophil activity modulation. Myeloperoxidase is a very potent enzyme of neutrophil primary granules, it participates in their oxidative burst and is able to oxidise biomolecules with or without an attachment to the neutrophil extracellular traps. Besides neutrophils, 5-hydroxytryptamine released from activated platelets also occurs at a site of inflammation. Microenvironmental pH is an important feature for myeloperoxidase activity, it changes its catalytic cycle and therefore an interaction with 5-hydroxytryptamine. At phagosomal pH 5-hydroxytryptamine decreased chemiluminescence and acted as an antioxidant. On the other hand, at physiological pH chemiluminescence was dose dependently increased upon 5-hydroxytryptamine concentration, 5-hydroxytryptamine acted as a myeloperoxidase substrate in this case. We noticed similar effect using N-acetyl-5-hydroxytryptamine. Decreased chemiluminescence was detected using isolated neutrophils in interaction with 5-hydroxytryptamine in the highest concentration used (10 µM) suggesting its antioxidant properties. In the following experiments we succeeded in induction of neutrophil extracellular traps and in visualisation of DNA structure with myeloperoxidase and elastase using confocal microscopy. To determine whether 5-hydroxytryptamine influences formation of extracellular traps, method of myeloperoxidase oxidation of 3,3´,5,5´-tetramethylbenzidine was used. This method suggested a decrease in structure formation, however, this finding needs further examination. This thesis demonstrates pH dependent interaction of 5-hydroxytryptamine with myeloperoxidase. 5-hydroxytryptamin acted as an antioxidant (at phagosomal pH) and as an alternative substrate (at physiological pH), and therefore can modulate neutrophil activity mediated by myeloperoxidase.
