Molekulární mechanismy účinků platinových chemoterapeutik u nádorových buněk tlustého střeva

Molecular mechanisms of the action of platinum-based chemotherapeutic drugs in colon cancer cells

Abstract:

Platinové komplexy jsou stále hlavní volbou v léčbě nádorových onemocnění a jsou často podávány mnoha pacientům podstupujícím chemoterapeutickou léčbu. Hlavní limitací v současnosti používaných platinových chemoterapeutik jako cisplatina jsou jejich četné vedlejší účinky a vrozená nebo získaná rezistence u mnohých nádorů. Pro překonání těchto nevýhod je potřeba i) objasnit dosud nevysvětlené molekulární mechanismy působení v současnosti používaných platinových léčiv, ii) syntetizovat a charakterizovat nové platinové komplexy s lepšími protinádorovými vlastnostmi, a/nebo iii) navrhnout vhodné kombinace platinových léčiv s efektivními senzitizátory pro posílení smrti nádorových buněk, s minimálním dopadem na zdravou tkáň. Cílem předložené práce je přispět k lepšímu pochopení molekulárních mechanizmů působení platinového(II) komplexu cisplatiny a platinového(IV) komplexu LA-12 s adamantylaminovým ligandem a najít nové originální přístupy ke zcitlivění lidských nádorových buněk tlustého střeva k cytotoxickému působení těchto léčiv. Jako první jsme ukázali, že rosiglitazon, syntetický ligand receptorů aktivovaných peroxizómovými proliferátory (PPARγ), byl schopen potencovat smrt indukovanou látkou LA-12 u lidských nádorových buněk tlustého střeva. Kombinace těchto látek navozovala synergické cytotoxické efekty prostřednictvím posílení mitochondriální dráhy apoptózy, závisle na kaspazách, ale nezávisle na proteinech p53 a Chk2. Apoptóza indukovaná kombinací rosiglitazonu s LA-12 byla preferenčně indukovaná v G1 fázi buněčného cyklu a posílená u nádorových buněk deficientních pro PTEN. RNA interference pomocí specifické PPARγ siRNA významně neovlivnila buněčnou smrt indukovanou kombinovaným působením rosiglitazonu a LA-12. Aplikace kombinace těchto látek nevedla k posílení smrti buněk odvozených z lidské nenádorové epiteliální tkáně tlustého střeva. Nově jsme ukázali, že SCH900776, vysoce selektivní ATP kompetitivní inhibitor kinázy Chk1, posiluje cytotoxické účinky cisplatiny nebo LA-12 u lidských nádorových buněk tlustého střeva. Kombinované působení SCH900776 s cisplatinou nebo LA-12 specificky posílilo apoptózu nádorových buněk ve fázi G1/S a stimulovalo jejich únik z mitózy a senescenci. Odpověď na kombinované působení uvedených látek byla dále významně ovlivněná přítomností proteinů p21, p53 a PTEN. Buněčná smrt indukovaná SCH900776 a cisplatinou nebo LA-12 byla výrazně zpomalená v nepřítomnosti proteinu p53, zatímco u buněk deficientních pro p21 bylo protinádorové působení kombinace látek významně urychlené a spojené se stimulací apoptózy v průběhu celého cyklu včetně G2/M fáze. Kooperativní cytotoxické účinky SCH900776 a cisplainy/LA-12 byly navíc usnadněny v nepřítomnosti proteinu PTEN. U nádorových buněk s chybějícím proteinem p53 nebo p21 docházelo ke zvýšenému výskytu polyploidie a významnému potlačení senescence v porovnání s kontrolními buňkami. Naše výsledky ukázaly, že proteiny p21 a PTEN jsou důležitými faktory, které určují odpověd nádorových buněk na kombinované působení SCH900776 a cisplatiny nebo LA-12. Získané výsledky přinášejí nové poznatky nutné k objasnění molekulárních mechanizmů zcitlivění nádorových buněk tlustého střeva k cytotoxickému působení cisplatiny a LA-12, zejména s ohledem na zapojení mitochondriální dráhy apoptózy, regulaci buněčného cyklu a úlohy kinázy Chk1/2. Tyto poznatky mohou přispět k lepšímu pochopení protinádorových účinků platinových látek a jejich terapeutického potenciálu v léčbě nádorů.

Abstract:

Platinum complexes are still a mainstay of cancer therapy as numerous patients under chemotherapy frequentlly receive platinum-based compounds. However, the limits of platinum chemotherapeutics commonly used in clinic such as cisplatin include various side effects, an inherent cancer cell resistance or development of acquired resistance for many tumors over time. In order to overcome these disadvantages, there is a need i) to elucidate yet unknown molecular mechanism of the action of currently used platinum drugs, ii) to introduce and characterize novel platinum-based complexes with better anticancer properties, and/or iii) to obtain convenient combinations of platinum drugs with effective sensitizers in order to increase cancer cell death, with minimum impact on healthy tissues. The present work aimed to contribute to the better understanding of the molecular action of the platinum(II) complex cisplatin and a recently introduced platinum(IV) adamantylamine ligand-containing complex LA-12, and to find new original approaches how to sensitize human colon cancer cells to the cytotoxic effects of these drugs. We showed for the first time that rosiglitazone, a synthetic ligand of peroxisome proliferator-activated receptor (PPARγ), was able to potentiate the human colon cancer cell death induced by LA-12. The drug combination induced synergistic cytotoxic effects through enhancement of caspase- and mitochondria-dependent apoptosis, independently of p53 and Chk2 proteins. The apoptosis induced by the combination of rosiglitazone with LA-12 was preferentially induced from G1 phase of the cell cycle, and reinforced in PTEN-deficient cancer cells. The siRNA-mediated downregulation of PPARγ did not affect the stimulation of cell death induced by the drug combination. The drug combination also did not enhance apoptosis in the cells derived from normal colon epithelial tissue. Next, we newly showed that SCH900776, a highly selective ATP competitive inhibitor of checkpoint kinase 1 (Chk1), potentiated the cytotoxic effects of cisplatin or LA-12 in human colon cancer cells. The combined treatment with SCH900776 and cisplatin or LA-12 apparently increased the G1/S phase-related apoptosis, and stimulated mitotic slippage and senescence of colon cancer cells. The response to the drug combinations was significantly affected by the cellular p21, p53, and PTEN status. The cell death induced by SCH900776 and cisplatin or LA-12 was significantly delayed in the absence of p53, but the anticancer action of the drug combinations was significantly accelerated in p21-deficient cells, which was associated with stimulation of apoptosis beyond G2/M cell cycle phase. The cooperative killing action of the drug combinations in HCT116 cells was further facilitated in the absence of PTEN. In contrast to their wt counterparts, the p53- or p21-deficient cells treated with SCH900776 and cisplatin or LA-12 could enter mitosis and became polyploid, and the senescence phenotype was strongly suppressed. Our results showed that p21 or PTEN proteins might serve as important determinants of cancer cells response to the combined action of SCH900776 and cisplatin or LA-12. The original results obtained within this work provide novel insights in the molecular mechanisms of colon cancer cell sensitization to the cytotoxic action of cisplatin and LA-12, with a particular focus on mitochondria-related apoptotic signaling, cell cycle regulation and the role of Chk1/2. These findings could contribute to the better understanding of the anticancer properties and future therapeutic potential of these platinum compounds.
