Abstract:

Cílem práce bylo zjištění vitality osiva řepky ozimé (dále jen řepky) různými laboratorními metodami a konfrontovat je s hodnotami polní vzcházivosti. Polní pokus probíhal v polních podmínkách na dvou lokalitách, a to v Chlumci nad Cidlinou a v Opavě, kde byla zjišťována polní vzcházivost. Laboratorní pokus probíhal ve třech variantách s použitím roztoku polyethylen glykolu (PEG 6000), a to na úrovni stresu -0,3, -0,5 a -0,7 MPa. V první variantě byla použita klíčidla, která byla na úrovni dvou stresu suchem, a to -0,5 a -0,7 MPa a kontroly s destilovanou vodou za standardní teploty. Pokus byl vyhodnocován ve třech termínech. Další varianta probíhala v Petriho miskách při dvou úrovních sucha, a to -0,3 a -0,5 MPa, kdy byly měřeny délky kořenů u klíčenců a povrch kořene klíčenců. Tato varianta byla umístěna do klimaboxu za teploty 30 °C, s následným vyhodnocením ve dvou termínech. Třetí varianta pokusu probíhala v Petriho miskách naplněných sterilním pískem, do kterého se následně nalil polyethylen glykol ve dvou koncentracích pro stres suchem, a to -0,3 a -0,5 MPa, a při teplotě 35 °C. Vyhodnocení proběhlo ve třech termínech. Pro následné vyhodnocení výsledků byla použita metoda výpočtu korelativního vztahu a analýza rozptylu. V této práci vyšlo, že mezi odrůdami je statisticky významný rozdíl u všech koncentrací ve všech pokusech. Byly zjištěny korelační vztahy jak v rámci pokusů, tak i mezi pokusy.

Abstract:

The goal of this thesis was to determine the vigor of rape seed by various methods. And then to compare the results of those with the field emergence. The experiment was conducted at field conditions and at two localities, those were Chlumec nad Cidlinou and Opava. The emergence of all varietes was determined here. The field experiment had three repeats. Laboratory experiments were conducted next, here the vigor of rape seed was determined. The experiment had three variants with the usage of polyethylene glycol (PEG 6000). The tests were conducted at three levels of stress, the levels were -0,3; -0,5; -0,7 MPa. The first method used the gemination box, that was the method with distilled water as control and -0,5; -0,7 levels of stress in standard temperature. This variant was evaluated three times. Next variant was conducted in petri dishes with -0,3; -0,5, where the length of roots and the area of seedlings was measured. This variant was placed in clima box at the temperature of 30 °C. This variant was evaluated two times. The third variant was conducted in petri dishes, which were filed with sterile sand. Which was then filled with PEG 6000 in two concentrations for drought stress, the levels of stress were -0,3; -0,5, this was once again placed in the clima box, and the temperature was 35 °C. This variant was evaluated three times. Then the method of corelative analysis and ANOVA were used for overall evaluation. The result was that there were statistically major differences between the varieties in all concentrations and all experiments. According to corelative analysis there is a clear relation within a in between the experiments. Also, according to this analysis, the strongest relation is between the Laboratory experiments and field emergence is the screening of seedlings.
