Abstract:

První část práce je zaměřena na studium nanočástic a vývoj insekticidů založených na nanočásticích. Navazuje na závěry studie benchmarkingu dvou různých přístupů pro automatickou detekci a identifikaci škůdců. Výsledkem studie byl autonomní robot, který na základě počítačového vidění a algoritmů byl nejen schopen detekovat patogen a identifikovat škůdce, ale také eliminovat škůdce aplikací insekticidů. Proto se první část práce zaměřuje na vliv nanočástic na rostliny a patogeny, které mohou být v budoucnosti považovány za novou generaci insekticidů. Byl studován vliv nanočástic ZnO, SiO2 NPs a Ag NPs a jejich toxicita na Tenebrio molitor a Solanum lycopersicum. Na základě biochemických analýz bylo zjištěno, že ZnO NPs nevykazovaly toxicitu u rostlin. Zároveň vybrané NPs prokázaly toxicitu na larvy potemníka moučného. Druhá část práce je věnována studiu ABC transportérů a umlčování těchto transportérů pomocí RNA interference. Bylo použito osm různých genů a dvě kontroly (ds-Actin jako pozitivní a ds-LacZ jako negativní kontrola). In vitro syntetizovaná dsRNA byla aplikována do kněžice zeleninové (Nezara viridula L.). Umlčování ABCH transportérů pomocí RNAi u N. viridula ukázalo, že Snu je vysoce konzervovaný gen a je nezastupitelný. Mortalita byla spočítaná pomocí vzorce Schneider-Orelli a exprese genů pomocí qPCR analýzy. Po deseti dnech od aplikace dsRNA prokázal pouze gen Snu vysoké procento úmrtnosti (víc než 90 %), zatímco všechny ostatní geny ABCH vykazovaly úmrtnost miň než 30 %.

Abstract:

The first part of the thesis focuses on the study of nanoparticles and the development of insecticides based on nanoparticles. It follows up on the conclusions of a benchmarking study of two different approaches to automatic pest detection and identification. The result of the study was an autonomous robot which, based on computer vision and algorithms, was not only able to detect the pathogen and identify pests, but also to eliminate pests by applying insecticides. Therefore, the first part of the thesis focuses on the effect of nanoparticles on plants and pathogens that may be considered in the future as a new generation of insecticides. The effect of ZnO, SiO2 and Ag nanoparticles were studied and their toxicity to Tenebrio molitor and Solanum lycopersicum. Based on biochemical analyses, ZnO NPs were not found to be toxic to plants. At the same time, selected NPs showed toxicity to the mealworms larvae. The second part of the thesis is devoted to studying ABC transporters and silencing them with RNA interference. Eight different genes and two controls were used (ds-Actin as positive and ds-LacZ as negative control). In vitro synthesized dsRNA, was applied to stink bug (Nezara viridula L.). Silencing ABCH transporters with RNAi in N. viridula has shown that Snu is a highly preserved gene and is necessary. Mortality was calculated using the Schneider-Orelli formula, and gene expression using qPCR analysis. Ten days after application of dsRNA, only Snu gene showed a high percentage of mortality (more than 90 %), while all other ABCH genes showed a mortality of less than 30 %.
