Abstract:

Tématem této diplomové práce zpracovávané formou rešerše je funkce a metabolismus sacharidů se zaměřením na člověka. V první části práce se zabývám obecným postavením sacharidů ve stravě a jejich biochemií, tedy strukturou a vlastnostmi. Tato část je velmi důležitá pro pochopení role sacharidů v lidském organismu a uvědomění si jejich nezaměnitelnosti. Hlavní část práce je rozdělená na tři celky, první se zabývá glukózou, nejdůležitějším sacharidem lidského organismu, a jeho metabolismem. V této části je podrobně rozebrána glykolýza, její enzymy, průběh, regulace a odbourávání jejího produktu. Dále se věnuji mechanismu glukoneogeneze a jeho srovnání s obrácenou glykolýzou. Druhou hlavní část práce tvoří informace o zásobním polysacharidu glykogenu a jeho metabolismu, tedy glykogeneze a gykogenolýza. Ve třetí části se pak věnuji hormonům ovlivňujícím metabolismus sacharidů, konkrétně glukagonu, jeho působení, metabolismu a regulaci sekrece a v neposlední řadě také inzulínu. Kapitola o inzulínu pojednává mimo jiné o vzniku, sekreci a metabolismu inzulínu, o inzulínových receptorech, intracelulární inzulínové kaskádě, translokaci GLUT4 transportérů a samozřejmě také o účincích inzulínu a jejich možném ovlivnění. Závěrečná část práce je věnována přehledu nemocí a chorob, které jsou způsobené poruchou metabolismu sacharidů. Zde se zaměřuji například na hyperglykémii, diabetes mellitus a jeho typy, hypoglykémii, glykogenózy, ale také poruchy metabolismu fruktózy, galaktózy a laktózy. Tento přehled chorob není kompletní, spíše má upozornit na různorodost poruch a tím také na důležitost role sacharidů v lidském těle.

Abstract:

The aim of my master thesis, written as a literary rewiew, is function and metabolism of carbohydrates, focusing on humans. The first part deals with the general status of carbohydrates in the diet and their biochemistry, ie structure and properties. This part is very important for understanding the role of carbohydrates in the human body and realizing their uniqueness. The main part is divided into three parts, the first is dealing with glucose, the most important carbohydrate in human body and its metabolism. There is examined in detail glycolysis, the enzymes, process, controling and degradation of the product in this section. Then I describe the mechanism of gluconeogenesis and its comparison with reverse glycolysis. The second part consists of the main storage polysaccharide glycogen metabolism and, therefore, glycogenesis and glycogenolysis. The third part is concentrated to hormone affecting carbohydrate metabolism, specifically glucagon, its function, metabolism, and regulation of secretion and, ultimately, insulin. The chapter on insulin deals with formation, secretion and metabolism of insulin, the insulin receptors, intracellular insulin cascade, translocation of GLUT4 transporters and, of course, on the effects of insulin and their possible interaction. The final part of the work is aimed to an overview of illnesses and diseases that are caused by disorder of carbohydrate metabolism. And there, for example, I describe hyperglycemia, diabetes mellitus and its types, hypoglycemia, glycogenosis, but also fructose, galactose and lactose metabolic disorders. This list of diseases is not complete, but rather to highlight the diversity of disorders and thus also the important role of carbohydrates in human body.
