Cloning, production and characterization of Photorhabdus spp. lectins

Abstract:

Lektiny jsou proteiny se schopností vázat sacharidy. Hrají roli v různých fyziologických i patologických procesech, jako je buněčná signalizace a komunikace, imunita, nádorová onemocnění, či adheze patogenu. Právě funkce lektinů jako mediátorů adheze patogenu k cílové tkáni je středem pozornosti, protože tento krok je v mnoha případech nezbytný pro zahájení infekce. Lektiny jsou tedy atraktivním terapeutickým cílem. Vzhledem ke vzrůstající míře rezistence k různým antibiotikům je velmi žádoucí nalezení alternativní léčby. Terapie cílená na lektiny může v budoucnosti pomoci při léčbě antibiotiky, případně antibiotika zcela nahradit. Tato diplomová práce je zaměřena na lektiny pojmenované PHL2 a PHL3 z bakterie Photorhabdus asymbiotica, lidského i hmyzího patogenu. Teoretická část práce obsahuje rešerši věnovanou historii, funkci a aplikacím lektinů. Krátce je také představen rod Photorhabdus se zaměřením na P. asymbiotica. Experimentální část popisuje produkci rekombinantních lektinů a jejich strukturně-funkční charakterizaci za použití široké škály molekulárně biologických, biochemických a biofyzikálních metod.

Abstract:

Lectins are ubiquitous sugar-binding proteins serving many purposes in both physiological and pathological processes. They play key role in cell signaling and communication, immunity, cancer, pathogen adhesion, etc. Involvement of lectins in pathogen adhesion is of particular interest, since this step might be crucial for the infection initiation. Owing to rise in antibiotic resistance, this marks adhesive lectins as an interesting future target for therapeutic intervention. Such therapy could complement or replace antibiotic treatment. This diploma thesis focuses on lectins denoted PHL2 and PHL3 from Photorhabdus asymbiotica bacterium, a dualistic pathogen infecting insect and human. Theoretical part of the thesis will review history, function and applications of lectins, and will briefly introduce Photorhabdus genus, with focus on P. asymbiotica. Experimental part will describe production of recombinant lectins and characterization of their structure and function. Array of molecular biological, biochemical and biophysical methods will be used to study these aspects.
