Abstract:

Kometová analýza je mikroelektroforetická metoda pro zjištění poškození DNA na úrovni jednotlivých buněk. Jedním z cílů této práce bylo zavedení kometové analýzy na MU LF Brno, přičemž byla použita alkalicko-neutrální modifikace, barvení stříbrem a hodnocení pomocí počítačového programu LUCIA. V rámci validace metody byl testován vliv emprie hodnotitele a typu softwaru (LUCIA, CASP) na získané výsledky. Bylo zjištěno, že v případě vhodně stanoveného experimentálního bodu je vliv zkušenosti na celkové výsledky minimální. Ovlivnění výsledků vybraným softwarem však může být v určitých případech značné. Optimalizovaná metoda kometové analýzy byla použita při zkoumání poškozujícího a protektivního vlivu vybraných přírodních látek na DNA leukemických buněk linie HL-60 a melanomových buněk linie A375. Studován byl poškozující vliv benzofenanthridinových alkaloidů chelerythrinu a sanguinarinu na molekulu DNA. Jak chelerythrin, tak sanguinarin indukovaly poškození DNA buněk HL-60 a míra poškození se zvyšovala s rostoucí koncentrací alkaloidů. Na buněčné linii HL-60 byly zkoumány protektivní účinky antioxidantů bajkalinu a kyseliny kávové. Jako kontrolní antioxidant sloužil trolox a oxidačním agens v těchto experimentech byl peroxid vodíku. Všechny použité antioxidanty výrazným způsobem redukovaly poškození DNA v případě oxidačního stresu vyvolaného peroxidem vodíku. Preinkubací s výše uvedenými antioxidanty byla zkoumána možnost, že při působení alkaloidů (3 μg/ml) je oxidační stres jedním z mechanismů, které se podílejí na poškození DNA buněk HL-60. Protektivní vliv antioxidantů vůči působení alkaloidů byl u chelerythrinu prokázán. Kyselina kávová však snižovala poškození DNA výrazně méně než trolox a bajkalin. V případě sanguinarinu nebyl ochranný vliv antioxidantů před poškozením DNA prokázán. Antioxidant bajkalin v kombinaci se sanguinarinem dokonce poškození navyšoval a to statisticky významně. Odlišná míra poškození DNA oběma alkaloidy byla pozorována i na buněčné linii A375 v závislosti na inkubaci. U adherentních buněk bylo poškození DNA chelerythrinem srovnatelné s kontrolním vzorkem, avšak u buněk v suspenzi bylo poškození chelerythrinem vyšší než v případě sanguinarinu. I když jsou zde studované alkaloidy strukturně velmi podobné, výsledky naznačují, že způsob jakým poškozují DNA studovaných buněčných linií je odlišný.

Abstract:

Presented dissertation thesis studies the effect of some benzophenanthridine alkaloids on different cell lines (HL-60, A375). Selected alkaloids were chelerythrine and sanguinarine. DNA damage was assessed by comet assay. The alkaline-neutral modification of comet assay was used throughout this study. This modification has advantage over widely used alkaline modification in being less demanding while still maintaining high sensitivity. The staining of comets was performed by silver staining, and comets evaluated by software applications LUCIA. Both alkaloids in all tested concentrations (1/0,65; 3; 6,5 and 10 μg/ml) were found to be toxic by means of DNA fragmentation (HL-60 culture) assessed by comet assay. Statistically there were no differences between chelerythrine and sanguinarine in 3 μg/ml concentration, while in concentration 6,5 μg/ml sanguinarine was more DNA damaging than chelerythrine. Contrary, in the highest concentration (10 μg/ml) chelerythrine has bigger impact on DNA fragmentation and was found more toxic than sanguinarine in the same concentration. According to published literature one of the possible mechanisms how benzophenanthridine alkaloids damage DNA is a production of reactive oxygen species. Therefore selected antioxidants were used as protective agents against DNA damaging alkaloids. In pilot experiments with hydrogen peroxide it was found that all antioxidants used lowered the oxidative damage although in different levels. The best one was trolox, followed by caffeic acid and baicalin, although the caffeic acid alone had relatively high damaging effect. The DNA damage by chelerythrine was ameliorated by all antioxidants, but the effect of caffeic acid was weak, hence it was not used in experiments with sanguinarine. The protective effect of antioxidants was not observed when using sanguinarine as a damaging agent, even generally potent trolox was not able to reduce the DNA damage. Experiments with cell line A375 showed that damage depended on procedure how the cells were incubated. As A375 is not a suspension living cell culture it has to be disaggregated by trypsinization before experiments. Trypsinization before incubation with alkaloids led to the increase of DNA damage ("trypsin effect"), while trypinization after incubation meant generally less fragmented DNA. However the effect of both alkaloids was not comparable. Results show that in some circumstances chelerythrine can be more genotoxic (contrary to generally published results) and the mechanism affecting DNA damage differs between studied alkaloids.
