The importance of population-wide genomic studies of genetic variability based on massive parallel sequencing technology and its impact on clinical practice

Abstract:

Tato práce je přehledem dokončených i nedokončených genomických studií a projektů variability populací. S rozvojem technologií sekvenace došlo k výraznému nárůstu v počtu genomických studií, které se snaží vytvořit referenční databáze pro genetické varianty v jednotlivých populacích. Vytvoření těchto databází by mohlo vést k lepšímu porozumění různým onemocněním, lepší interpretaci sekvenačních dat a tím k rozvoji personální medicíny. V první části práce jsou stručně popsány metody paralelního sekvenování a nejčastěji používané přístroje v těchto projektech. Hlavní část práce je pak zaměřena především na sumarizaci výsledků jednotlivých studií, ať už multipopulačních či lokálních. Závěr práce je zaměřen spíše na význam poznatků z těchto studií a jejich potencionální využití v klinické praxi a personální medicíně.

Abstract:

This thesis is an overview of both finished and unfinished genomic studies and projects of population variability. With technology development came a singnificant increase in number of genomic studies, which are trying to create a refrence database for genetic variants in individual populations. The creation of these databases could lead to better understanding of certain diseases, better intepretation of sequence data and therefore to development of personal medicine. Methods of parallel sequencing and the most used instruments in these projects are briefly described in the first part of the thesis. The main part of the thesis is then focused on summarizing results of individual studies whether multi-populational or local. The end of the thesis is focused rather on the significance of the findings from these studies and their potential use in clinical practise and personal medicine.
