Structural-functional basis of psychrotolerance of haloalkane dehalogenase enzyme isolated from extremophilic organism

Abstract:

Halogenalkandehalogenasy jsou mikrobiální enzymy, které katalyzují hydrolýzu vazby uhlík-halogen v halogenovaných alifatických uhlovodících, přičemž se uvolňuje halogenidový iont, příslušný alkohol a proton, jako reakční produkty. Halogenalkandehalogenasy katalyzují reakci s velkým environmentálním a biotechnologickým významem a potenciálním využitím v bioremediaci, biosenzorech, dekontaminaci vojenských látek, syntéze opticky čistých sloučenin, v buněčném zobrazování a značení proteinů. Efektivní využití biokatalyzátorů v těchto aplikacích často vyžaduje dostupnost enzymů se speciálními vlastnostmi pro výrobní podmínky. Tyto vlastnosti mohou být získané izolací nových biokatalyzátorů z extremofilních organizmů, které produkují enzymy zvané extremozymy. Tyto enzymy jsou přirozeně schopné přežít extrémní podmínky, jako je vysoká nebo nízká teplota, vysoké a nízké pH, vysoký tlak nebo vysoká salinita. Tato diplomová práce se zabývá identifikací, izolací a biochemickou charakterizací nového enzymu DpcA z organizmu Psychrobacter cryohalolentis K5, který představuje prvního psychrofilního člena rodiny halogenalkandehalogenas.

Abstract:

Haloalkane dehalogenases are microbial enzymes which catalyze the hydrolysis of a carbon-halogen bond in halogenated aliphatic hydrocarbons, releasing a halide ion, a corresponding alcohol and a proton as the reaction products. HLDs catalyze the reactions of great environmental and biotechnological significance with potential application in the bioremediation, the biosensing, the decontamination of warfare agents, the synthesis of optically pure compounds, the cellular imaging and the protein tagging. The effective use of biocatalysts in these applications requires availability of enzymes with specific properties under process conditions. Such properties can be obtained by isolation of novel biocatalysts from the extremophiles, producing the enzymes called extremozymes. These enzymes are able to operate under extreme conditions such as high or low temperature, high or low pH, pressure or high salinity. This Diploma Thesis describes identification, isolation and biochemical characterization of a novel haloalkane dehalogenase DpcA from Psychrobacter cryohalolentis K5, representing the first psychrophilic member of the haloalkane dehalogenase family
