Determination of metallothionein in earthworms Eisenia foetida after exposure to metal nanocompounds

Abstract:

Diplomová práce se zabývá problematikou nanomateriálů, především jejich vlivem na životní prostředí. Teoretická část je věnována výhodám využívání nanočástic, ale také riziky s nimi spojenými a interakcemi nanočástic s buňkami. Pozornost je zaměřena na nanočástice TiO2 a ZnO. Další část se týká biomarkerů a s nimi souvisejícím oxidačním stresem, celkovou antioxidační kapacitou, multibiomarkerovou odezvou a významnými biologickými markery metallothioneiny, superoxiddismutázou, katalázou a glutathionem. Praktická část je zaměřena na metodiku stanovení hladiny biomarkeru metallothioneinu pomocí enzymové imunoanalýzy ELISY. Cílem praktické části je stanovení hladiny biomarkeru metallothioneinu ve tkáních modelového organismu Eisenia foetida po expozici nanočásticemi TiO2 a ZnO.

Abstract:

The diploma thesis deals with the issue of nanomaterials, especially their impact on the environment. The theoretical part is devoted to the advantages of using nanoparticles, but also the risks associated with them and the interactions of nanoparticles with cells. Attention is focused on ZnO and TiO2 nanoparticles. The next part concerns biomarkers and related oxidative stress, total antioxidant capacity, multibiomarker response and important biological markers metallothionein, superoxide dismutase, catalase and glutathione. The practical part is focused on the methodology of determining the level of the biomarker metallothionein using the enzyme-linked immunosorbent assay ELISA. The aim of the practical part is to determine the level of metallothionein biomarker in the tissues of the model organism Eisenia foetida after exposure to TiO2 and ZnO nanoparticles.
