Characterization of the function of the proteins involved in the telomere length regulation

Abstract:

Telomeres are complex structures composed of DNA and proteins located at the ends of linear chromosomes . They are important in protecting the ends of chromosomes, ensuring that they are distinguished from double-stranded DNA breaks. Telomerase enzyme, which consists of two subunits: telomerase RNA (TR) and telomerase reverse transcriptase (TERT), is involved in maintaining telomere length. If the enzyme telomerase is not active, telomere shortening and cellular senescence can be activated. In addition to telomerase, however, there are a number of proteins that provide telomere protection. For example, in animals, the telomere-associated protein complex is called Shelterin. Proteins that are similar to Shelterin proteins have recently been described in plants. In this work I deal with the telomere-binding protein TRB (Telomere repeat binding) from the plant Arabidopsis thaliana. This protein belongs to the Single myb histone (SMH) family of proteins and is believed to be involved in both telomeric terminal maintenance and regulation of nonelomeric processes. The aim of this work was to cross protein mutant plants that are associated with telomere length regulation. The purpose of crossing was to obtain double mutants for attrb3-attert, which are then suitable for more detailed analysis of processes associated with regulation of telomeric functions in A. thaliana. Another aim was to confirm mutual protein-protein interactions between AtKu70/80 proteins, AtTRB proteins and AtTRBP, which were described as being associated with the telomeric sequence, by a yeast two-hybrid system.

Abstract:

Teloméry sú komplexné štruktúry zložené z DNA a proteínov nachádzajúce sa na koncoch lineárnych chromozómov. Sú dôležité pri ochrane koncov chromozómov, zabezpečujú ich odlíšenie od dvojvláknových zlomov DNA . Na udržiavaní dĺžky telomér sa podieľa enzým telomeráza, ktorý je tvorený dvomi podjednotkami: telomerázovou RNA (TR) a telomerázovou reverznou transkriptázou (TERT). Ak enzým telomeráza nie je aktívna, dochádza ku skracovaniu telomér a môže byť aktivovaná bunková senescencia. Okrem telomerázy však existuje celá rada proteínov, ktoré sa podieľajú na udržiavaní a ochrane telomér. Napríklad u živočíchov sa komplex proteínov asociovaných s telomérami nazýva Shelterin. U rastlín boli v nedávnej dobe popísané proteíny, ktoré sú podobné proteínom z komplexu Shelterin. V tejto práci sa zaoberám telomér-väzbovým proteínom TRB (Telomere repeat binding) z rastliny Arabidopsis thaliana. Tento proteín patrí do Single myb histone (SMH) rodiny proteínov a predpokladá sa, že sa podieľa ako na udržiavaní telomerických koncov tak aj na regulácii netelomerických procesov. Cieľom tejto práce bolo kríženie rastlín mutantných pre proteíny, ktoré sú spojené s reguláciou dĺžky telomér. Účelom kríženia bolo získanie dvojitých mutantov v génoch attrb3-attert, ktorí sú následne vhodní pre detailnejšie analýzy procesov spojených s reguláciou telomerických funkcií u A. thaliana. Ďalším cieľom bolo overenie vzájomných proteín-proteínových interakcií medzi proteínmi AtKU70/80, AtTRB a AtTRP1, ktoré boli popísané ako asociované s telomerickou sekvenciou, využitím kvasinkového dvojhybridného systému.
