Electrochemical biosensor for detection of methylated DNA

Abstract:

Bakalářská práce se v teoretické části zabývá problematikou epigenetických změn a jejísouvislostí s nutrigenomikou. Největší pozornost je věnovaná problematice metylované DNA,která je nejčastější epigenetickou změnou DNA. Pozornost je věnovaná také možným chybámv metylaci DNA, jakož to původcům metabolických, kardiovaskulárních a karcinogenníchonemocnění. Na základě současných poznatků jsou v této práci prezentovány nejčastějšímetody detekce metylované DNA, především detekce pomocí biosenzorů aelektrochemických metod. Poslední část teoretické práce se pak věnuje samotné problematicebiosenzorů, jejich stavby, rozdělení a modifikací.Praktická část se zaobírá návrhem a sestavením biosenzoru na metylovanou DNA, od volbypřevodníku až po správnou modifikaci. Následně je popsaná charakterizace a ověřenífunkčnosti biosenzoru na metylovanou DNA. V poslední části jsou prezentovány výsledkytěchto měření a jejich zhodnocení.

Abstract:

The theoretical part of the bachelor thesis deals with the problematics of epigenetic changesand their connection with nutrigenomics. The problematics that is given the most attention ismethylated DNA, which is the most common epigenetic change. Possible errors in DNAmethylation are also mentioned here, as they are the cause of cancer and various metabolicand cardiovascular diseases. The most frequently used methods of methylated DNA detectionare described in this thesis, primarily focusing on detection by biosensors and electrochemicalmethods. The last part of the theoretical part deals with the problematics of biosensors, theirconstruction, division, and modification.The practical part is aimed at the process of design and assembly of a biosensor formethylated DNA, from the choice of a converter to correct modification. The characterisationof the biosensor and verification of the functionality is then described. In the last part, theresults of those measurements and their evaluation is presented.
