Role of HMGB proteins in telomere biology

Abstract:

Cílem této diplomové práce bylo objasnit vliv proteinů HMGB1 na stabilitu chromatinu v lidských buňkách. Stabilita chromatinu se projevuje také na úrovni konců chromozomů, všechny provedené analýzy měly tedy souvislost s těmito oblastmi - telomerami. Byla měřena jejich délka, míra zastoupení tzv. subtelomerických oblastí (TERRA transkripty) a také exprese a aktivita podjednotek enzymu, který telomery syntetizuje (telomeráza). Doplňujícím experimentem byla analýza exprese genu pro protein HMGB1, který odhalil zastoupení tohoto proteinu ve sledovaných buňkách. Kromě neošetřených buněk linie MFC-7 byly použity buňky transfekované shRNA, která měla způsobit umlčení proteinu HMGB1, a také buňky transfekované nespecifickým konstruktem shRNA, u kterých se nepředpokládal specifický efekt na expresi proteinu a linie sloužila jako kontrola pro postižení případného ovlivnění sledovaných parametrů procesem tranfekce. U obou transfekovaných linií byla pozorovaná snížená aktivita telomerázy, zkrácení telomer i menší zastoupení TERRA transkriptů, avšak snížená exprese podjednotek telomerázy pozorována nebyla. Analýza exprese HMGB1 ukázala, že ke specifickému umlčení proteinu HMGB1 nedošlo, zatímco v buňkách transfekovaných nespecifickým konstruktem došlo ke zvýšení jeho exprese až na 450 % jeho hladiny ve standardních buňkách MCF-7. Ačkoli cíle diplomové práce byly formálně splněny ve všech bodech, pro analýzu role HMGB1 v oblasti udržování telomer uvedenými technikami bude zapotřebí nově připravených buněčných linií. Nicméně zjištěné výsledky ukazují na fakt, že proces transfekce u lidských buněk linie MCF-7 významně ovlivňuje regulaci exprese genů pro telomerázy a má vliv na stabilitu telomer.

Abstract:

This diploma thesis´ aim was to study the influence of the HMGB1 protein on chromatin stability in humans. Since the stability of chromatin is linked with the ends of chromosomes, all the anaysis done were related with these parts of chromosomes - telomeres. Their length was measured, the subtelomeric regions were analyzed and also the expression of the subunits of the enzyme which synthesizes telomeres. An additional experiment was the analysis of the expression level of HMGB1 protein in cell lines used. Apart from the line MCF-7 without any transfection the cell line transfected with shRNA which was supposed to silence HMGB1 protein was used and a cell line transfected with nonspecific shRNA. It was shown that in both transfected cells the telomerase activity was lower, telomeres were shorter, the cells contained less TERRA transcripts, but the expression of the telomerase subunits was the same or even higher. But it was not due to the lower expression of HMGB1 protein as originally supposed according to the previous studies, because the analysis of the expression level of HMGB1 protein revealed that the transfection with shRNA had not been sucsessful in its significant silencing. There was the same level of HMGB1 protein in silenced line compared with the line without any transfection (MCF-7). Moreover, in the control cell line which was nonspecifically transfected, the protein expression was 4.5 times higher. Although all the goals of the diploma thesis were reached, another kind of source material is needed to reveal the specific effect of HMGB1 protein to telomeres and telomerase activity. However, the results show a significant effect of transfection procedure itself to the processes of regulation of gene expression and telomere maintenance.
