The role of plants in treatment of neurodegenerative diseases

Abstract:

Identifikace rostlinných produktů potencionálně terapeuticky využitelných při léčbě neurodegenerativních onemocnění (jako např. Alzheimerova choroba, Parkinsonova choroba, ALS) je v dnešní době stále značným vědeckým zájmem. Důvody jsou poměrně zřejmé, neboť počet pacientů postižených některým z NDO se neustále zvyšuje, velká část z těchto onemocnění má časté genetické podmínění a konvenční léčba bývá problematická či naprosto nedostačující z důvodu vážných vedlejších účinků. Tato práce se v rešeršní části zabývá principem a klasifikací neurodegenerativních disfunkcí se zaměřením na Alzheimerovu chorobu, popisem a rozdíly mezi konvenční a alternativní léčbou a v závěru charakteristikou vybraných 8 peruánských rostlinných druhů se slibnou predispozicí jako terapeutického prostředku pro léčbu NDO. V druhé části práce jsou tyto vybrané vzorky podrobeny laboratorní zkoušce inhibice enzymu acetylcholinesteráza. Konkrétně byly zkoušce podrobeny extrakty rostlinných druhů @Alternanthera brasiliana@ (f. @Amaranthaceae@), @Alternanthera flavescens@ (f. @Amaranthaceae@), @Ayapana triplinervis@ (f. @Asteraceae@), @Dysphania ambrosioides@ (f. @Amaranthaceae@), @Eleutherine bulbosa@ (f. @Iridaceae@), @Mansoa alliacea@ (f. @Bignoniaceae@), @Petiveria alliacea@ (f. @Petiveriaceae@), @Phyllanthus niruri@ (f. @Phyllanthaceae@) ve srovnání s konvenčně využívanými léčivy galathaminem a neostigminem. Bohužel i přes velmi slibný potenciál těchto vzorků odpovídaly všechny výsledné hodnoty IC50 koncentracím vyšším než 512 μg/ml. Ve srovnání s konvenčními inhibitory nebyla tato interakce dostačující, neboť jejich reakční koncentrace dosáhly řádově nižších výsledků. Galanthamin vykazoval výslednou hodnotu pro IC50 2,347 μg/ml a neostigmin 1,268 μg/ml. Výsledky vyjadřují vysokou specifitu enzymu AChE a s tím související náročnost hledání aktivních přírodních látek, které by byly schopny tento enzym inhibovat.

Abstract:

The identification of plant products with potential therapeutically use in the treatment of neurodegenerative diseases (e.g. Alzheimer's disease, Parkinson's disease, ALS) is currently of interest in scientific circles. The reason being the ever-increasing number of patients with NDOs. Given the genetic causation of these diseases, conventional treatment can be problematic or completely insufficient due to the serious side effects.In the research part of this study, the principle and classification of neurodegenerative dysfunctions with a focus on Alzheimer's disease are discussed, also description and differences between conventional and alternative treatment are presented and finally the characteristics of 8 the selected Peruvian plant species with a promising predispositions as a therapeutic agent for NDO are included. In the second part of this study, these samples are tested in a laboratory experiment for inhibition acetylcholinesterase enzyme. The sampled plants were namely @Alternanthera brasiliana@ (f. @Amaranthaceae@), @Alternanthera flavescens@ (f. @Amaranthaceae@), @Ayapana triplinervis@ (f. @Asteraceae@), @Dysphania ambrosioides@ (f. @Amaranthaceae@), @Eleutherine bulbosa@ (f. @Iridaceae@), @Mansoa alliacea@ (f. @Bignoniaceae@), @Petiveria alliacea@ (f. @Petiveriaceae@), @Phyllanthus niruri@ (f. @Phyllanthaceae@) and they were compared with conventionally used medication galathamine and neostigmine. Unfortunately, all the results of IC50 corresponded to concentrations higher than 512 μg / ml despite the very promising potential of all the samples. This interaction was not sufficient compared to the conventional inhibitors. Their active concentrations achieved range lower results. Galanthamine showed a final IC50 2.347 μg/ml and neostigmine 1.268 μg/ml.The results express the wide uniqueness of the AChE enzyme and the consequential difficulties of finding natural active species that would be able to inhibit this enzyme.
