Abstract:

Tato práce se zabývá návrhem, syntézou a studiem fotoindukovaných "click" a "unclick" reakcí p-hydroxyfenacyl (pHP) derivátů. pHP byla nejprve testována jako fotoodstupitelná chránící skupina pro 1,2,3-triazoly. Na základě úspěšného fotochemického odchránění bylo testováno použití pHP skupiny jako fotolabilní spojky. Nakonec byla provedena one-pot reakce, která umožňuje selektivně spojit a rozpojit dvě molekuly in situ pomocí světla o různých vlnových délkách. V prvním kroku reaguje pHP azid s derivátem cyklooktynu fotochemicky generovaném z příslušného cyklopropenonu za vzniku derivátu 1,2,3-triazolu. V následujícím kroku je tento konjugát rozštěpen účinkem UV záření. Fotochemické vlastnosti pHP derivátů 1,2,3-triazolů (absorpční spektra, kvantové výtěžky fotoodstoupení, acidobazické vlastnosti) byly detailně studovány. Vyvinutá procedura "catch & release" má široké možnosti využití v molekulární biologii a materiálové chemii.

Abstract:

This work deals with the design, synthesis and study of photoinduced "click" and "unclick" reactions of the p-hydroxyphenacyl (pHP) derivatives. First, the photorelease of 1,2,3-triazole from a pHP photoremovable protecting group was examined. Based on the successful photochemical deprotection, the use of pHP moiety as a photolabile linker was explored. Finally, a one-pot reaction, which enables to selectively connect and disconnect two molecules, was performed in situ by light at two different wavelengths. The system is based on the fotochemical generation of a derivative of cyclooctyne from the corresponding cyclopropenone and its click reaction with pHP azide to form 1,2,3-triazole in the first step. This conjugate is cleaved by UV irradiation in the subsequent step. Photochemical properties (absorption spectra, quantum yields of deprotection, acidobasic properties) of the derivatives of pHP 1,2,3-triazoles were studied. The developed "catch & release" procedure has broad applications in molecular biology and material chemistry.
