Abstract:

Studie poskytuje souhrn popisů třinácti dobře popsaných druhů hub dřevomorek, ve vztahu k nejdestruktivnějšímu druhu Serpula lacrymans. Jsou zmíněny historie jejich poznání, názory na jejich původ, výskyt, biochemické vlastnosti, možnosti vizuální identifikace, preparaci a mikroskopování, možnosti odběru vzorků i možnosti molekulární detekce, které se vztahují zejména k druhu S. lacrymans. Molekulárně genetické metody mohou poskytnout téměř stoprocentní spolehlivost při identifikaci organizmů. V této souvislosti jsou popsány možnosti molekulární detekce, včetně nově vyvinutého originálního postupu detekce S. lacrymans metodou využívající PCR s druhově specifickými primery zacílenými do genu pro velkou podjednotku ribozomální RNA v kombinaci s restrikční analýzou produktu. Závěrem je pojednáno o možnostech sanace a případného využití enzymatického potenciálu těchto hub v průmyslu a farmacii.

Abstract:

This study provides a summary of descriptions of thirteen species of fungi taxonomically closely related to domestic dry rot fungus, Serpula lacrymans, in relation to this species as the most destructive representative of the group. The history of the study of this group as well as its possible origin, abundance, biochemical features, methods of visual identification, preparation and microscopic observation, sampling methods and possibilities of the molecular detection of S. lacrymans are also mentioned. Methods of molecular genetics may provide almost absolute reliability of the detection of organisms. This text thus describes a newly developed original method of the detection of S. lacrymans exploiting PCR with species-specific primers directed to the ribosomal RNA large subunit gene in combination with restriction analysis of the product. Finally, the possibilities of the remediation of the objects damaged by the fungus and potential industrial and pharmacological use of fungal enzymes are tracted.
