MicroRNAs as a disease-modulating factor in skin diseases in humans

Abstract:

MikroRNA (miRNA) se významným způsobem podílejí na regulaci velkého množství buněčných procesů, přičemž její deregulace může vést ke vzniku různých patologií. Tato práce v úvodní části shrnuje současné poznatky o biogenezi miRNA a mechanismech, kterými miRNA post-transkripčně reguluje genovou expresi v buňkách. Další část popisuje některé možnosti detekce miRNA ze vzorku, čehož se využívá ke stanovení expresních profilů miRNA, které jsou charakteristické pro jednotlivé orgány, či dokonce samotné buňky v normálním i patologickém stavu. Identifikace jednotlivých miRNA je nutná k porozumění jejich funkce v organismu. Dále tato práce tedy popisuje expresi a funkci několika známých miRNA, které se významně podílejí na ontogenezi zdravé kůže a které zásadně ovlivňují funkci imunitního systému. Tyto poznatky pak lze využít k popisu role jednotlivých miRNA ve vzniku a průběhu kožních onemocnění u člověka, což je hlavním cílem této práce. Konkrétně tato práce diskutuje podíl miRNA na vzniku patologií u psoriasis vulgaris, atopické dermatitidy, sklerodermie, dermatomyositis a maligního melanomu, kde je na závěr zmíněno i potenciální využití miRNA v diagnostice, prognostice a terapii těchto onemocnění.

Abstract:

MicroRNAs (miRNAs) plays an important role in the regulation of a large number of cellular processes, while their deregulation may lead to various pathologies. In this thesis, the introduction summarizes the current understanding about miRNA biogenesis and the mechanisms by which miRNAs exert post-transcriptionally their effects on gene expression. The following part of the thesis describes several possibilities for detecting miRNAs within a sample, which can then be used to determine the expression profiles of miRNAs that are characteristic for individual organs or tissues. Identification of individual miRNAs is essential for proper understanding of their particular function within an organism. The thesis then describes the expression profiles and functions of certain miRNAs that are known to play a significant role in the ontogeny of healthy skin and that are relevant for immune functions. This knowledge can then be used to describe the role of individual miRNAs in the development and progress of skin diseases in humans; this being the main goal of this thesis. In particular, this thesis discusses the role of miRNA molecules in pathogenesis of psoriasis vulgaris, atopic dermatitis, scleroderma, dermatomyositis, and malignant melanoma. In the final part of the thesis, the potential applications of miRNA for diagnostics, prognostics and therapy of the above diseases is illustrated.
