Role of Wnt signaling in ovarian cancer progression

Abstract:

Ovariální karcinom je jedním z nejfatálnějších maligních onemocnění u žen. V pokročilých stádiích je často spojen s akumulací maligního výpotku – ascitu v peritoneální dutině, který pacientkám znepříjemňuje již natolik komplikované onemocnění. Pro výzkumné účely ovšem znamená vysoce informativní stadium malignity s relativně snadným přístupem. Faktory pro-nádorového mikroprostředí, které ascites poskytuje diseminovaným nádorovým buňkám, například extracelulární vezikuly, totiž velmi významně ovlivňují rychlost progrese. Role, kterou extracelulární vezikuly hrají v progresi nádoru, šíření a odolnosti vůči terapii, nedávno otevřela nové cesty v základním i translačním výzkumu mnohých malignit. Zde sledujeme roli transportu proteinů signalizace Wnt na extracelulárních vezikulech a jiných částicích v ascitu pacientek s ovariálním karcinomem. Klíčové aspekty progrese ovariálního karcinomu související s aberantní signalizací Wnt jsou totiž mnohem méně pro-zkoumané než u jiných malignit. Naše výsledky poukazují na ligand Wnt5a, který byl nejčastěji zastoupen ve frakcích různých ascitů na exozomech a lipoproteinových částicích s vysokou hustotou. Této analýze předcházela validace specifity komerčně dostupných protilátek Wnt, při které jsme zjistili většinovou nespolehlivost daných protilátek. Pro odhalení mechanismů příspěvku ascitického mikroprostředí k progresi ovariálního karcinomu jsme dále ukázali možnost odvození nového modelového systému v oblasti výzkumu ovariálního karcinomu – nádorové organoidy z maligního ascitu. Na tomto modelu jsme prokázali přítomnost epitelových, mezotelových a možných nádorových kmenových buněk, čímž jsme provedli první kroky v charakterizaci těchto nově odvozených komplexních struktur. Další směry výzkumu buněk tvořících nádorové organoidy, stejně jako buněk a acelulárních faktorů maligního ascitu, včetně signálních molekul, budou vést k lepšímu pochopení mechanismu regulace metastatické kolonizace a poskytnou zase o něco pokročilejší možnost predikce chování maligních buněk v těle pacientek s ovariálním karcinomem.

Abstract:

Ovarian cancer is one of the most fatal malignancies in women. In advanced stages, it is often associated with the accumulation of malignant effusion - ascites in the peritoneal cavity, which makes patients even more uncomfortable with such a complicated disease. For research purposes, however, it represents a highly informative stage of malignancy with relatively easy access. Indeed, the pro-tumor microenvironment factors that ascites provides to disseminated tumor cells, e.g extracellular vesicles, significantly affect the rate of progression. The role that the extracellular vesicles play in tumor progression, spread, and resistance to therapy has recently opened up new avenues in the basic and translational research of many malignancies. Here, we investigate the role of Wnt signaling protein transport on extracellular vesicles and other particles in the ascites of patients with ovarian cancer. Indeed, key aspects of ovarian cancer progression associated with aberrant Wnt signaling are much less studied than in other malignancies. Our results point to the ligand Wnt5a, which was most often present in the fractions of various ascites on exosomes and high-density lipoprotein particles. This analysis was preceded by validation of the specificity of commercially available Wnt antibodies, and we found the majority unreliability of the antibodies. To reveal the mechanisms of the contribution of the ascitic microenvironment to the progression of ovarian cancer, we have further shown the possibility of deriving a new model system in the field of ovarian cancer research – the tumor organoids from malignant ascites. In this model, we demonstrated the presence of epithelial, mesothelial and possible tumor stem cells, that taking the first steps in the characterization of these newly derived complex structures. Further research with tumor organoid-forming cells, as well as cells and acellular malignant ascites factors, including signaling molecules, will lead to a better understanding of the mechanism of metastatic colonization and provide a slightly more advanced way to predict malignant cell behavior in ovarian cancer patient
