Physiological and Pathological consequences of cellular senescence

Abstract:

Buněčná senescence je stav trvalého zastavení buněčného cyklu, který je doprovázen morfologickými, biochemickými i molekulárními změnami, přičemž buňka jako taková zůstává stále metabolicky aktivní. Jde o významný nádorově supresorový mechanismus, který se podílí na regeneraci tkání a účastní se embryonálního vývoje. Se zvyšujícím se věkem dochází k akumulaci senescentních buněk ve tkáních, neboť je imunitní systém není schopen dostatečně rychle odstranit. V takovém případě senescence paradoxně podporuje vznik patologických stavů, jakými jsou rakovina a s věkem související onemocnění. Cílem této práce je popsat fyziologické i patologické důsledky buněčné senescence.

Abstract:

Cellular senescence is a state of permanent cell cycle arrest, which is accompanied by morphological, biochemical and molecular changes, wherein the cell itself remains still metabolically active. It is an important tumor suppressor mechanism that is involved in regeneration of tissues and embryonic development. Increasing age leads to accumulation of senescent cells in tissues, because the immune system is unable to eliminate them fast enough. In this case, senescence paradoxically supports development of pathological conditions such as cancer and age-related diseases. The aim of this thesis is to describe the physiological and pathological consequences of cellular senescence.
