Antimicrobial peptides and their effect on wound healing.

Abstract:

Problémem západního světa je častý výskyt chronických ran, z nichž některé jsou také infikovány multirezistentními bakteriemi. Vhodným kandidátem na léčení ran s rezistentními bakteriemi jsou antimikrobiální peptidy. Teoretická část práce popisuje biologii kůže, proces hojení rány a charakterizuje chronickou ránu či vliv bakterií na hojení ran. Poslední kapitola je věnována katelicidinům s vlivem na hojení rány. Cílem této práce je popis problematiky hojení ran a potenciálu peptidů – katelicidinů tyto rány účinně hojit. Dále je cílem navrhnout mechanismus inteligentního uvolňování katelicidinů pro odstranění problémů bránící klinické aplikaci těchto látek. V praktické části je testována antibakteriální aktivita katelicidinů proti kožním patogenům s rezistencí na antibiotika a řešen problém ztráty aktivity peptidu katelicidinu LL-37. Katelicidiny inhibovaly růst multirezistentních kožních patogenů Acinetobacter baumannii a Pseudomonas aerugunisa. Taktéž je testováno inteligentní uvolňování katelicidinů in vitro či in vivo v rámci animálního experimentu. Inteligentní uvolňování katelicidinů představuje funkční cestu pro aplikaci peptidů do komplikovaných ran. Nicméně do budoucna bude potřeba koncept inteligentního uvolňování ještě více rozvíjet a testovat.

Abstract:

Frequent occurrence of chronic wounds, some of which infected with multidrug-resistant bacteria, represents a serious problem in the Western world. A suitable candidate for the treatment of wounds with resistant bacteria includes antimicrobial peptides. The theoretical part of the thesis deals with the biology of the skin and the wound healing process, focusing on characteristic features of chronic wounds and the influence of bacteria on wound healing. The last chapter is devoted to cathelicidins with their effect on wound healing. The aim of the present thesis is to describe the wound healing process and the potential of peptides–cathelicidins–to effectively heal wounds. Furthermore, the aim is to propose a mechanism of intelligent release of cathelicidins to eliminate the problems hindering the clinical application of these agents. The practical part of the thesis discusses the antibacterial activity of cathelicidins against skin pathogens with antibiotic resistance, and the problem of loss of activity of the cathelicidin LL-37 peptide. Cathelicidins inhibited the growth of multidrug-resistant skin pathogens Acinetobacter baumannii and Pseudomonas aerugunisa. In an animal experiment, the intelligent release of cathelicidins in vitro or in vivo is also tested. Intelligent release of cathelicidins represents a functional route for peptide application to complicated wounds. However, there is no doubt, the concept of smart release will need to be further developed and tested in the future.
