Limiting and inhibiting factors of bacterial inorganic sulfur compounds oxidation

Abstract:

Práce se zabývá různými aspekty biooxidace síry bakterií Acidithiobacillus ferrooxidans. Výsledky mají souvislost s činností bakterie v biotechnologii a biogeochemických procesech. V růstové kultuře byla studována variabilita vybraných kinetických konstant biooxidace síry, jež zásadně ovlivňují celý bioproces: výtěžek biomasy na síře (YX/S) a maximální rychlost oxidace síry (v). Výsledky získané z mnoha kultur a během dlouhého období prokázaly vysokou variabilitu těchto konstant, bez vzájemné korelace. Příčina není známa, tento výsledek se však úzce týká konstrukce kinetických modelů bakteriální oxidace síry a vede k obezřetnosti v pohledu na obecnou platnost těchto konstant. Bakterie vyvíjejí aktivitu často i v nerůstových podmínkách, kdy biooxidace síry odpovídala kinetice Michealise a Mentenové. Zde se variabilita „biokonstant“ studovala u Michaelisovy konstanty pro síru (Km) a maximální rychlosti oxidace síry. Bylo zjištěno, že Km je závislá na koncentraci buněk a fyziologickém stavu bakterií, oproti tomu reálná oblast pH a teplot nemá na Km zásadní vliv. Část disertační práce se zabývá aplikací komerčního kitu na stanovení ATP v kultuře At. ferrooxidans oxidující síru jako míry růstu biomasy a oxidace substrátu. Luminiscenční reakcí byl určen buněčný obsah ATP u bakterie At. ferrooxidans ve vztahu k parametrům růstu buněk a oxidace síry a byly postiženy změny buněčného ATP v různých fázích kultury. Byly eliminovány inhibiční vlivy prostředí na luciferasu a byl určen maximální obsah ATP v buňkách, jenž je interpretován v souvislosti s postulovanou limitací sírou. Dokumentovaly se fáze dobré korelace intenzity bioluminiscence s rychlostí růstu buněk a oxidace síry, jakož i fáze, kde tato korelace neplatí, neboť se objevil efekt „odlišného metabolického módu“. Pro význam thiosíranu jako sirného meziproduktu se studovala interakce thiosulfátdehydrogenasy jako enzymu s potenciální aktivitou v biooxidaci síry. Byla provedena afinitní chromatografie thiosulfátdehydrogenasy z bakterie At. ferrooxidans. Ligandem byl imobilizovaný cytochrom c. Nová laboratorně připravená matrice založená na zesíťovaném triazinu (TAT) s imobilizovaným cytochromem c tedy reprezentuje použitelnou metodu i pro purifikaci thiosulfátdehydrogenasy.

Abstract:

This dissertation thesis describes various aspects of elemental sulfur oxidation by Acidithiobacillus ferrooxidans. The results are related to the activity of the bacteria in biotechnology and biogeochemical processes. The purpose of this study was to investigate variability in elemental sulfur metabolism using selected kinetic constants: growth yield on sulfur (YX/S) and the maximum rate of the sulfur oxidation (v). Results obtained from many cultures showed a high variability of these constants, without correlation. The reason is unknown, but this result is closely related to the construction of kinetic models of bacterial sulfur oxidation and leads to a cautionary view of the general validity of these constants. Bacteria developed activity often in non-growing conditions, where the elemental sulfur oxidation corresponded to Michealis-Menten kinetics. The variability of the Michaelis constant for sutur (Km) and maximum rate of sulfur oxidation was studied. It was found that Km is dependent on cell concentration and cell storage length. In contrast, pH and temperature did not represent factors influencing Km. Part of the thesis deals with the application of a commercial kit for the ATP determination in the sulfur-oxidizing culture of At. ferrooxidans. Cellular ATP content of At. ferrooxidans was determined with a bioluminescence assay in relation to batch growth and oxidation of elemental sulfur. Changes in cellular ATP were monitored at various stages of culture. Inhibitory effect of the environment on luciferase was eliminated and also the maximum cell content of ATP was determined. Good correlation phases of intensity bioluminiscence with cell growth rate and sulfur oxidation were documented, and the phase where the correlation is not valid because " different metabolic mode " occurred. The interactions of thiosulfate dehydrogenase as a potential enzyme activity in the elemental sulfur oxidation were studied because of possibility of thiosulfate to be a sulfur intermediate. intermediate. Affinity chromatography of thiosulfate dehydrogenase of At. ferrooxidans with cytochrome c as a ligand was performed. A new laboratory-prepared matrix based on crosslinked triazine with immobilized cytochrome c represented an applicable method for thiosulfate dehydrogenase purification.
