The role of TRAMP complex in transposition of Ty1 elements in Saccharomyces cerevisiae

Abstract:

Ty1 transposony jsou mobilní genetické elementy kvasinek, které jsou strukturně i funkčně blízké retrovirům. K transposzici těchto elementů však dochází velmi zřídka, a to i přesto, že Ty1 RNA je jednou z nejhojnějších mRNA v buňce, kde tvoří až 0,8% celkové RNA. Dosud nebylo objasněno, jak buňka reguluje transpozici Ty1 elementů, ale bylo identifikováno mnoho kvasinkových genů účastnících se tohoto procesu. Cílem této práce bylo objasnění role komplexu TRAMP v transpozici Ty1 elementů. Tento komplex je zásadní komponentou kontroly kvality RNA v jádře kvasinek a skládá se z nekanonické poly(A) polymerasy Trf4/Trf5, zinkového proteinu Air1/Air2 a RNA helikasy Mtr4. TRAMP komplex je schopný rozpoznat vadnou molekulu RNA, označit jí oligo(A) koncem a tím umožnit její degradaci exosomem, makromolekulárním komplexem s 3´− 5´ exonukelasovou aktivitou. Bylo ukázáno, že delece TRF4 a TRF5 genů, jakožto i delece genu kódující exonukleasovou podjednotku exosomu, RRP6, způsobí narušení standardních hladin Ty1 RNA. V této práci jsme otestovali, zda narušené hladiny Ty1 RNA v kmenech mutantních v TRAMP či exosom komplexu, ovlivňují četnost transpozicí Ty1 elementů. Zjistili jsme, že četnost transpozice v těchto kmenech je výrazně snížena. Opětovně jsme otestovali hladiny Ty1 RNA a nepozorovali jsme jejich narušení v daných mutantech. Naši pozornost jsme tedy zaměřili na další krok retrotranspozice, translaci. To ukázalo, že hladiny Ty1 strukturního proteinu, TyA, jsou výrazně sníženy ve všech námi testovaných mutantních kmenech.

Abstract:

Ty1 transposons are mobile genetic elements which are structurally and functionally related to retroviruses. Despite the fact that Ty1 RNA is one of the most abundant mRNA species in yeast (up to 0,8 % of total RNA), their transposition occurs at very low rate. Multiple regulators and their role in metabolism of Ty1 elements were indentified, but general mechanism regulating retrotransposition have not been described yet. We are particularly interested in the role of TRAMP complex in the process of retrotransposition. The TRAMP complex is responsible for nuclear RNA processing, surveillance and degradation and consists of the non-canonical poly(A) polymerase Trf4p/Trf5p, the Zn-knuckle protein Air1p/Air2p and the RNA helicase Mtr4p. The role of TRAMP complex is to target RNA substrate by adding oligo(A) tail and thus promoting its degradation by exosome, a multiprotein complex with 3´− 5´ exonuclease activity. It has been reported, that the levels of Ty1 sense and antisense transcripts are altered in trf4 and trf5 mutant strains as well as in exosome mutant, rrp6. Here, we aimed to investigate whether altered RNA levels of either sense or antisense Ty1 transcripts are reflected in the retrotransposition rate. We observed that mobility of the Ty1 elements is significantly decreased in these strains. In order to find which step of Ty1 transposition is affected in TRAMP and exosome mutants, we examined Ty1 transcript abundance, protein levels and cDNA levels. We observed that levels of Ty1 structural protein, TyA, are significantly decreased in all mutant strains in contrast to the unaltered levels of Ty1 mRNA.
